چکیده

The automated rrvethod of O’Neal and Gochman [C/in. Chem. 16, 985 (1970)] for determination of serum and urine amylase activityhas been modified to increase its accuracy, precision, and efficiency. Sampling rate is 50 specimens per hour. The analysis depends on ct-amylase hydrolyzing glycogen to maltose-like subunits, which are measured with the copper neocuproine reaction. Results obtained by this method are compared to those for a manual reference method [C/in. Chem. 6, 434 (1960)].

An automated, saccharogenic method for determining serum amylase activity.

An automated adaptation of the Somogyl saccharogenic determination of serum amylase is described in which conventional AutoAnalyzer modules are used. Adequate sensitivity with short incubation is achieved by incorporating glucose oxidase and catalase in the substrate to destroy serum glucose during incubation. Maltose and other dialyzable oligosaccharides are measured with the alkaline copper-n...

New saccharogenic determination of alpha-amylase in serum and urine.

I describe a new kinetic enzymatic saccharogenic method for assaying alpha-amylase in human serum and urine. alpha-Amylase liberates maltose from starch. This is successively acted on by alpha-glucosidase, mutarotase, and glucose dehydrogenase. The resulting conversion of NAD+ to NADH, measured at 340 nm, during a 20-min incubation reflects amylase activity. Endogenous glucose is destroyed befo...

Clinical significance of urinary amylase in diagnosis of pancreatic disease.

Studies were made on the methods for urinary amy lase determination, stability of the enzyme, isoamylase in urine, normal ranges, diagnostic value in various diseases (pancreatic and non-pancreatic), and relationship between urinary amylase excretion and other laboratory tests. The saccharogenic method of Somogyi was chosen after comparing the three methods of Somogyi, Wohlgemuth, and Van Loon....

A Study of the Starch-iodine Complex: a Modified Colorimetric Micro Determination of Amylase in Biologic Fluids.

The limitations, inaccuracies, and practical difficulties of saccharogenic methods are discussed. A modified colorimetric microdetermination of amylase is described in which the digestion of starch is measured by the decrease in the starch-iodine color. Experimental data show that there are two other serum factors that can also cause a fall-off in color: (1) an immediate 10-15% depression of co...

Factitiously low amylase values.

When a commercial substrate was used in the determination of serum amylase activities of patients with acute pancreatitis, results much higher than those reported by the manufacturer with similar patients were found. A failure to quantitate all of the reducing groups liberated during incubation may account for the lower results. A modified saccharogenic method has been developed, which, by incr...
