چکیده

Various chemical compounds have been described to induce photosensitization of tumor cells resulting in cell death. We studied the effect of merocyanine-540 (MC-540) on both leukemic and normal bone marrow (BM) cells. Acute promyelocytic leukemia (HL-60) and common acute lymphoblastic leukemia antigen-positive nun-1, non-B acute lymphoblastic leukemia (Reh) cell lines were incubated with MC-540 and simulta neously exposed to white light. Normal human BM and mixtures of leukemic cells with BM cells were treated under similar conditions. At constant illumination rates of 50,000 Ix, significant (at least 4 to 5 logs) tumor cell destruction was obtained with MC-540 concentrations of 20 «ig/mlor more for HL-60, and 10 MÕ/nil or more for Reh cells. Incubation of BM under equivalent conditions preserved 18.0% of granulocytemacrophage colony-forming units and 14.2% of erythroid burst-forming units. Similar results were obtained when tumor cells were mixed with irradiated BM and then treated with MC-540. In summary, cell photosensitization with MC-540 has a selective cytotoxic effect towards leu kemic cells and therefore may be useful for purging tumor cells from

Limiting-dilution analysis for the determination of leukemic cell frequencies after bone marrow decontamination with mafosfamide or merocyanine 540.

To stimulate a leukemia remission marrow, cell suspensions of normal human bone marrow were mixed with human acute lymphoblastic or myelogenous leukemic cells of the CCRF-SF, Nalm-6, and K-562 lines. The cell mixtures were incubated in vitro with mafosfamide (AZ) or with the photoreactive dye merocyanine 540 (MC-540). A quantity of 10(4) cells of the treated suspensions was dispensed into micro...

Assessment of Cytotoxicity of Dimethyl Sulfoxide in Human Hematopoietic Tumor Cell Lines

Background: Dimethyl Sulfoxide (DMSO) is a solvent most broadly used as a cryopreservative agent. Antitumor effects of DMSO is a recently recognized phenomenon. In this study, cytotoxic effects of DMSO on human monocytes and T leukemic cell lines has been investigated in vitro. Methods: Human leukemic T cells (Molt-4 and Jurkat) and monocytes (U937 and THP1) were cultured in complete RPMI medi...

Cytotoxicity of Metoprolol on Leukemic Cells in Vitro

Background: β-Blockers have shown considerable cytotoxic, anti-tumor and anti-angiogenic effects. Metoprolol, a β-Blocker with anti-inflammation, anti-tumor and anti-angiogenic properties has been widely used for treatment of some cardiovascular diseases such as angina, hypertension, heart failure and myocardial infraction. Limited data exist about the cytotoxic effects of metoprolol on human c...

Sensitivity of Human Leukemic Cells to Carvedilol

Background and Objective: Carvedilol, a non-specific β-blocker, has been used for treatment of hypertension, stroke and congestive heart failure. The therapeutic effects of β blockers in cancer patients have been shown. Carvedilol has considerable anti-inflammatory, anti-tumor and anti-angiogenic properties. In this study, the effects of carvedilol on proliferation of human U937 and Molt-4 leuk...

THE EFFECT OF HYPERTHERMIA ON THE DIFFERENTIATION OF LEUKEMIC CELL LINES

Treatment of human promonocytic leukemic cell line U937 with mild hyperthermia in the temperature range of 40-43°C resulted in differentiation of these cells into monocyte/macrophage-like cells in a heat dose and time dependent manner. This process was accompanied by marked morphological, functional and proliferational changes. U937 cells which normally grow in supension in the logarithmic...
