Thrombomodulin in human gestational tissues: placenta, fetal membranes and myometrium.

چکیده
PURPOSE In intervillous space, thrombomodulin (TM) can be a key component of the protein C anticoagulant pathway that controls the balance between coagulation and anticoagulation/ /fibrinolysis via activation of protein C (APC). In our working hypothesis we assume that not only TM from the placenta, but also TM from myometrium might be engaged in this protective mechanism. To determine whether this is potentially possible, we decided to measure TM concentration in placenta and myometrium, and also in fetal membranes. MATERIAL AND METHODS The study group consisted of 35 parturient women who delivered at term by cesarean section. Strips of placenta, fetal membranes and myometrium, as well as venous blood samples were collected during operation. The tissues were homogenized. TM was measured by immunoenzymatic method (ELISA). The concentration of TM antigen in placenta was 18.76 +/- 3.83 ng/mg proteins, in fetal membranes 8.57 +/- 1.64 ng/mg proteins and in myometrium 4.72 +/- 1.93 ng/mg proteins, while in blood plasma it was 0.063 +/- 0.016 ng/mg proteins. CONCLUSIONS It was shown for the first time that thrombomodulin is present in gestational myometrium and fetal membranes. The results support the hypothesis that not only placental TM, but also myometrial TM can participate in maintaining the fluidity of the blood in utero-placental circulation.

Parathyroid hormone-related protein (PTHrP) mRNA splicing and parathyroid hormone/PTHrP receptor mRNA expression in human placenta and fetal membranes.
During human pregnancy, parathyroid hormone-related protein (PTHrP) and parathyroid hormone (PTH)/PTHrP receptor are produced by the uterus, placenta, fetal membranes (amnion and chorion) and developing fetus. PTHrP alternative 3' mRNA splicing results in transcripts which encode three PTHrP isoforms and have been identified in amnion. Uteroplacental PTHrP expression is greatest in amnion and i...
متن کاملMarkers of protein synthesis are increased in fetal membranes and myometrium after human labour and delivery.
Preterm birth remains one of the leading causes of neonatal death. Inflammation and maternal infection are two of the leading aetiological factors for preterm birth. Labour is associated with increased production of proinflammatory cytokines, chemokines and prolabour mediators in human gestational tissues. In non-gestational tissues, synthesis of proinflammatory and prolabour mediators is regul...
متن کاملDietary Flavonoids as Therapeutics for Preterm Birth: Luteolin and Kaempferol Suppress Inflammation in Human Gestational Tissues In Vitro
Infection/inflammation is commonly associated with preterm birth (PTB), initiating uterine contractions and rupture of fetal membranes. Proinflammatory cytokines induce matrix metalloproteinases (MMPs) that degrade the extracellular matrix (ECM) and prostaglandins which initiate uterine contractions. Nuclear factor-κB (NF-κB) and activator-protein- (AP-)1 have key roles in the formation of thes...
متن کاملThe endogenous production of hydrogen sulphide in intrauterine tissues
BACKGROUND Hydrogen sulphide is a gas signalling molecule which is produced endogenously from L-cysteine via the enzymes cystathionine beta-synthase (CBS) and cystathionine gamma-lyase (CSE). The possible role of hydrogen sulphide in reproduction has not yet been fully investigated. It has been previously demonstrated that hydrogen sulphide relaxes uterine smooth muscle in vitro. The aim of the...
متن کاملThe Citrus Flavone Nobiletin Reduces Pro-Inflammatory and Pro-Labour Mediators in Fetal Membranes and Myometrium: Implications for Preterm Birth
Spontaneous preterm birth is the leading cause of infant death and of neurological disabilities in survivors. A significant proportion of spontaneous preterm births are associated with infection. Infection activates inflammation which induces a cascade of events that leads to myometrial contractions and rupture of fetal membranes. In non-gestational tissues, the citrus flavone nobiletin has bee...
