Sternbauer, K. 2005. Metabolic studies in cattle using the hyperinsulinemic euglycemic clamp technique. Doctoral dissertation. ISSN 1652-6880, ISBN 91-576-7037-4. Hyperinsulinemic euglycemic clamp, HEC, originally developed for determing insulinmediated glucose disposal and insulin sensitivity in humans, was used and evaluated in cattle. In the present study the following investigations were performed: the bovine physiological responses during drug-induced insulin resistance, the influence of liberal or restricted concentrate feeding on insulin sensitivity, the impact of two different physical activity levels on insulin sensitivity and muscle characteristics, and finally the expression of abomasal efflux rates in relation to different blood glucose levels. Clenbuterol and flumethasone treatment induced insulin resistance in calves lasting less than 17 hrs and 3 days post-treatment, respectively. The effect of flumethasone was more manifest than that of clenbuterol. Insulin sensitivity and muscle characteristics did not differ between one group of calves exposed to light exercise and another group of calves not exercised for 4 to 5 weeks. The results indicated that growth may have an influence on insulin sensitivity. The blood glucose levels of heifers fed concentrate at the upper limit, according to Swedish feeding standards, were higher than in heifers fed quantities at the lower limit. There was no change in insulin sensitivity or insulin levels between the two groups. A high plasma glucose level was shown to reduce the abomasal efflux rate and increase the abomasal pH in non-pregnant, non-lactating cows. The results show that the HEC technique may be a useful means for investigating druginduced insulin resistance and can also be used to create specific experimental conditions. Ruminant gluconeogenesis may still occur to some extent during HEC application, especially in lactating cows, and therefore this must be considered in future studies.

Comparison between the euglycemic-hyperinsulinemic clamp and the use of proxies for determination of insulin sensitivity in horses

Background Accurate quantification of insulin resistance is important for diagnosing and determining the efficacy of treatment in patients with equine metabolic syndrome (EMS). The euglycemic-hyperinsulinemic clamp (EHC) is a goldstandard method for measuring insulin sensitivity (IS) but the complexity of the technique limits its use in research. The use of proxies based on fasting values offer...

Undercarboxylated osteocalcin does not correlate with insulin resistance as assessed by euglycemic hyperinsulinemic clamp technique in patients with type 2 diabetes mellitus

UNLABELLED BACKGROUND Recent in vitro and in vivo studies have suggested a critical role of osteocalcin (OC), especially the undercarboxylated form (ucOC), in insulin secretion and insulin sensitivity. The objective of this study was to investigate the association between serum ucOC levels and insulin resistance in humans with type 2 diabetes mellitus. FINDINGS We measured serum ucOC level...

Insulin Sensitivity of Heifers on Different Diets

The hyperinsulinemic euglycemic clamp technique was used to investigate the effect on insulin sensitivity of 2 different diets used in practical cattle feeding in calves. Ten 4 to 5-month-old heifer calves were allocated to 2 feeding groups, LO or HI, to obtain growth rates of 400 g/day or 900 g/day. The heifers were fed and housed individually for 5 weeks. Growth rates close to calculated rate...

Comparison of the [13C]glucose breath test to the hyperinsulinemic-euglycemic clamp when determining insulin resistance.

OBJECTIVE With increasing emphasis on the recognition of the metabolic syndrome and early type 2 diabetes, a clinically useful measure of insulin resistance is desirable. The purpose of this study was to evaluate whether an index of glucose metabolism, as measured by (13)CO(2) generation from ingested [(13)C]glucose, would correlate with indexes from the hyperinsulinemic-euglycemic clamp. RES...

Insulin responses to selective arterial calcium infusion under hyperinsulinemic euglycemic glucose clamps: case studies in adult nesidioblastosis and childhood insulinoma.

Selective arterial calcium stimulation and hepatic venous sampling (ASVS) for insulin secretion is used as a diagnostic procedure in patients with insulinomas or adult nesidioblastosis. In some of those patients, severe hypoglycemia requiring urgent glucose administration occurs during the procedure. Such glucose administration, however, may affect the results and damage the validity of the tes...
