Self organization of Fibroblasts and Keratinocytes in morphogenesis of dermal-epidermal junction in human skin equivalents
نویسنده
چکیده
Introduction Dermal epidermal junction (DEJ) is a complex macromolecular structure which establishes the boundary between two major skin compartments, epidermis and dermis and the cellular components of these layers play a major role in its organization (1). We are interested in the structural and functional analysis of the epidermis formed in human skin equivalent (HSE) models that uses co-cultures of fibroblasts & keratinocytes (Threedimensional organotypic co-cultures) to derive the epidermal layer. Self-organizing capabilities and live cell fluorescence imaging of these two cell types in co-cultures on three-dimensional tissue constructs (dermal layer) will be used to determining the composition and conditions for the co-cultures that form epidermis structurally and functional similar to those in human skin.

In vitro Co-Culture of Human Skin Keratinocytes and Fibroblasts on a Biocompatible and Biodegradable Scaffold
Background: Extensive full-thickness burns require replacement of both epidermis and dermis. In designing skin replacements, the goal has been to re-create this model and make a product which has both essential components. Methods: In the present study, we developed procedures for establishing confluent, stratified layers of cultured human keratinocytes on the surface of modified collagen-chito...
متن کاملAging Alters Functionally Human Dermal Papillary Fibroblasts but Not Reticular Fibroblasts: A New View of Skin Morphogenesis and Aging
Understanding the contribution of the dermis in skin aging is a key question, since this tissue is particularly important for skin integrity, and because its properties can affect the epidermis. Characteristics of matched pairs of dermal papillary and reticular fibroblasts (Fp and Fr) were investigated throughout aging, comparing morphology, secretion of cytokines, MMPs/TIMPs, growth potential,...

Dermal Contributions to Human Interfollicular Epidermal Architecture and Self-Renewal
The human interfollicular epidermis is renewed throughout life by populations of proliferating basal keratinocytes. Though interfollicular keratinocyte stem cells have been identified, it is not known how self-renewal in this compartment is spatially organized. At the epidermal-dermal junction, keratinocytes sit atop a heterogeneous mix of dermal cells that may regulate keratinocyte self-renewa...
متن کاملBone marrow stromal cells, preadipocytes, and dermal fibroblasts promote epidermal regeneration in their distinctive fashions.
Mesenchymal cell types, under mesenchymal-epithelial interaction, are involved in tissue regeneration. Here we show that bone marrow stromal cells (BMSCs), subcutaneous preadipocytes, and dermal fibroblasts distinctively caused keratinocytes to promote epidermal regeneration, using a skin reconstruction model by their coculture with keratinocytes. Three mesenchymal cell types promoted the survi...
