چکیده

Transcription is defined as the synthesis of RNA from a DNA template. In bacteria, transcription of all genes is catalysed by a single RNA polymerase, whereas in the eukaryotic nucleus, three different RNA polymerases — Pol I, Pol II and Pol III — carry out the synthesis of different classes of RNA. Pol I and Pol III transcribe a limited number of genes encoding ribosomal RNAs, transfer RNAs and small nuclear RNAs. By contrast, Pol II is responsible for transcription of the thousands of protein-encoding genes, and does so using protein machinery comprising approximately 60 polypeptides with a combined molecular mass of more than 3 million daltons (MDa). Here, we describe the complex organization of these transcription factors and their various functions in directing Pol II to promoter DNA and in responding to the array of physiological and developmental cues that are essential for proper gene expression.

The Potential Mechanism of ZFX Involvement in Cell Growth

Background:The zinc-finger X linked (ZFX) gene encodes a transcription factor that acts as a regulator of self-renewal of stem cells. Due to the role of ZFX in cell growth, understanding ZFX protein-protein interactions helps to clarify its proper biological functions in signaling pathways. The aim of this study is to define ZFX protein-protein interactions and the role of ZFX in cell growth. ...

Eukaryotic and archaeal TBP and TFB/TF(II)B follow different promoter DNA bending pathways

During transcription initiation, the promoter DNA is recognized and bent by the basal transcription factor TATA-binding protein (TBP). Subsequent association of transcription factor B (TFB) with the TBP-DNA complex is followed by the recruitment of the ribonucleic acid polymerase resulting in the formation of the pre-initiation complex. TBP and TFB/TF(II)B are highly conserved in structure and ...

Transcription factors in DNA replication.

Accumulating evidence suggests the involvement of transcription factors in the regulation of DNA replication in eukaryotic cells. Almost all eukaryotic DNA viruses contain binding sites for transcription factors which function as auxiliary elements for DNA replication initiation at replication origins, and, indeed, the binding of transcription factors to these elements has been shown to stimula...

Same same but different: The evolution of TBP in archaea and their eukaryotic offspring

Transcription factors TBP and TF(II)B assemble with RNA polymerase at the promoter DNA forming the initiation complex. Despite a high degree of conservation, the molecular binding mechanisms of archaeal and eukaryotic TBP and TF(II)B differ significantly. Based on recent biophysical data, we speculate how the mechanisms co-evolved with transcription regulation and TBP multiplicity.

The σ enigma: Bacterial σ factors, archaeal TFB and eukaryotic TFIIB are homologs

Structural comparisons of initiating RNA polymerase complexes and structure-based amino acid sequence alignments of general transcription initiation factors (eukaryotic TFIIB, archaeal TFB and bacterial σ factors) show that these proteins are homologs. TFIIB and TFB each have two-five-helix cyclin-like repeats (CLRs) that include a C-terminal helix-turn-helix (HTH) motif (CLR/HTH domains). Four...

Evolution of eukaryotic transcription: insights from the genome of Giardia lamblia.

The Giardia lamblia genome sequencing project affords us a unique opportunity to conduct comparative analyses of core cellular systems between early and late-diverging eukaryotes on a genome-wide scale. We report a survey to identify canonical transcription components in Giardia, focusing on RNA polymerase (RNAP) subunits and transcription-initiation factors. Our survey revealed that Giardia co...
