The acetate activating enzyme of beef heart.
نویسنده
چکیده
Assay System-The assay system, previously described (1, 6, 7), is based on the conversion of acetyl CoA to citrate in the presence of malic dehydrogenase and condensing enzyme. The oxalacetate necessary for the condensation reaction is generated by the oxidation of malate in the presence of DPN, and the rate of DPN reduction is proportional to the concentration of AAE present, within the limits of the assay conditions (1). The specific activity is defined as micromoles of DPNH formed per mg. of protein per 15 minutes. A unit of enzyme activity is defined as that amount which leads to the formation of 1 ,uM of DPNH in 15 minutes. Preparation of Beef Heart Mitochondrial Particles-Fresh beef heart is freed from fat and connective tissue and diced. The subsequent steps are identical with those described previously for beef liver mitochondria (S), except that the acetone step is omitted and the particles are finally sus-

Effects of dietary lead acetate and aluminosilicates on the antioxidative defense system of broilers’ muscle tissues
The objective of this study was to evaluate the effect of dietary supplements with lead acetate and aluminosilicates (ATN) on antioxidative enzyme activities and lipid peroxidation of the heart and skeletal muscle tissues of broiler chickens. Broilers were allotted to four diets including the control group, the Pb group, the aluminosilicate-ATN (antitoxic nutrient) group and the Pb+ATN group, i...
متن کاملPurification and properties of tyrosine-activating enzyme of hog pancreas.
Activation reactions of this type are catalyzed by enzymes from various sources (2-6) and specific activating enzymes may be required for each amino acid (2). The first highly purified, activating enzyme was a tryptophan-activating enzyme from beef pancreas, studied by Davie et al. (7). The isolation and properties of a tyrosine-activating enzyme from hog pancreas (8) are described in this report.
متن کاملThe acetate activating enzyme system of heart muscle.
Nachmansohn and Machado (1) discovered a soluble enzyme system in extracts of brain tissue which catalyzed the formation of active acetate in the presence of ATPI and CoA. Active acetate was detected by the acetylation of choline. Later Lipmann (2, 3) reported an enzyme system in extracts of pigeon liver which catalyzed the CoA and ATP-dependent acetylation of sulfanilamide by acetate. The iden...
متن کاملThe comparison of growth, survival rate and behavior of Oscar fish (Astronauts ocellatus) fries fed by Artemia urmiana, earthworm and beef heart supplemented diets
This study was conducted to evaluate the effect of different fresh feed supplemented to commercial diet on growth and survival of Oscar fish (Astronauts ocellatus) (initial weight: 1.58-1.70 g) during 60-day period. The husbandry environment consisted of glass aquariums appropriate to growth with a photoperiod cycle of 12 L: 12 D. The water temperature was kept at 28 ±1°C. 360 fries of Oscar (i...
متن کاملPurification and properties of acetyl coenzyme A synthetase from bovine heart mitochondria.
Acetyl coenzyme A synthetase has been partially purified from many sources including yeast, bacteria, molds, plants, mammalian organs, and pigeon liver (5-11). Substrate amounts of this enzyme, partially purified by the method of Hele from beef heart mitochondria (lo), have been used for the isolation of acetyl adenylate from a reaction mixture containing all reactants except coenzyme A (4). Al...
