Microtubules turn over rapidly

چکیده
Microtubules turn over rapidly arc Kirschner recalls that the mid-1980s “was a very innovative time.” While the era was giving rise to stonewashed jeans and rap music, cell biology was entering new territory as it finally went molecular. For cell biologists who, like Kirschner, were interested in the cytoskeleton, there was an even more exciting possibility: the modification of interesting proteins to form reagents that could be used to follow dynamics inside of cells. Kirschner’s attention turned specifically to biotinylated tubulin. M

Direct observation of microtubule pushing by cortical dynein in living cells
Microtubules are under the influence of forces mediated by cytoplasmic dynein motors associated with the cell cortex. If such microtubules are free to move, they are rapidly transported inside cells. Here we directly observe fluorescent protein-labeled cortical dynein speckles and motile microtubules. We find that several dynein complex subunits, including the heavy chain, the intermediate chai...
متن کاملAssembly and turnover of detyrosinated tubulin in vivo
Detyrosinated (Glu) tubulin was prepared from porcine brain and microinjected into human fibroblasts and Chinese hamster ovary (CHO) cells. Glu tubulin assembled onto the ends of preexisting microtubules and directly from the centrosome within minutes of its microinjection. Incorporation into the cytoskeleton continued until almost all of the microtubules were copolymers of Glu and tyrosinated ...
متن کاملMicrotubules of the kinetochore fiber turn over in metaphase but not in anaphase
In previous work we injected mitotic cells with fluorescent tubulin and photobleached them to mark domains on the spindle microtubules. We concluded that chromosomes move poleward along kinetochore fiber microtubules that remain stationary with respect to the pole while depolymerizing at the kinetochore. In those experiments, bleached zones in anaphase spindles showed some recovery of fluoresce...
متن کاملFast Microtubule Dynamics in Meiotic Spindles Measured by Single Molecule Imaging: Evidence That the Spindle Environment Does Not Stabilize Microtubules
Metaphase spindles are steady-state ensembles of microtubules that turn over rapidly and slide poleward in some systems. Since the discovery of dynamic instability in the mid-1980s, models for spindle morphogenesis have proposed that microtubules are stabilized by the spindle environment. We used single molecule imaging to measure tubulin turnover in spindles, and nonspindle assemblies, in Xeno...
متن کاملDifferential turnover of tyrosinated and detyrosinated microtubules.
Turnover of tyrosinated and detyrosinated microtubules ([Tyr]MTs and [Glu]MTs, respectively) was analyzed by the combined use of hapten-mediated immunocytochemistry and peptide-specific antibodies. Cells were microinjected with hapten-labeled tubulin and then processed for triple-label immunofluorescence to determine the pattern of incorporation of the injected subunits into [Tyr]- and [Glu]-MT...
