Pili (fimbriae) of Branhameh Species
نویسنده
چکیده
PURPOSE: Pili (fimbriae) have frequently been found to be involved in the attachment of bacteria to mucosal epithelial cells, an important initial step in the disease process. The purpose of this study was to determine if BrunhameZZa catarrhalis expresses type 4 pili. MATERIALSANDMETHODS: Piliated B. caturrhalis phenotypic characteristics of colony morphology, agar corrosion, twitching motility, competence for deoxyribonucleic acid (DNA) transformation, autoagglutination, and pellical formation were observed. DNA was isolated from Branhamella spp. and used in genomic Southern hybridizations with a Moruxella bouis pilin gene as a probe. Electron microscopy of negatively stained bacteria was carried out to visualize pili. RESULTS: B. caturrhalis has several (but not all) of the phenotypic characteristics that are related to the presence of type 4 (MePhe) pili in closely related Moraxella spp., including competence for DNA transformation, autoagglutination, pellicle formation, colony morphology, and pitting of agar. The one phenotype we have not found that is generally characteristic of type 4 piliated bacteria is twitching motility. Genomic Southern hybridization analysis using a cloned M. bovis Q pilin gene as a probe reveals DNA homologous to the Q pilin gene in B. catarrhalis, Brunhamellu ovis, Branhamella caviae, and Branhamella cuniculi. Examination of B. catarrhalis strain ATCC25240 by electron microscopy reveals two different kinds of pili. One kind appears similar to other type 4 pili, whereas a second class is short pili extending outward from all portions of the bacteria. CONCLUSION: Phenotypic, electron-microscopic, and hybridization data are all consistent with type 4 pili being present on some B. catarrhalis strains.

In order to study the relationship between hrmolysin / ashesins of Escherichia coli and occurrence of urinary tract infection ( UTI), the capacity of 570 isolated E.coli in agglutination of human or guinea pig erythrocytes and their lysis were tested. These isolates were obtained from the urine of patients with bacteriuria and from the stool of healthy people. Three patterns of hemagglutinat...
متن کاملN. elongata Produces Type IV Pili That Mediate Interspecies Gene Transfer with N. gonorrhoeae
The genus Neisseria contains at least eight commensal and two pathogenic species. According to the Neisseria phylogenetic tree, commensals are basal to the pathogens. N. elongata, which is at the opposite end of the tree from N. gonorrhoeae, has been observed to be fimbriated, and these fimbriae are correlated with genetic competence in this organism. We tested the hypothesis that the fimbriae ...

Pseudomonas aeruginosa K/2PfS, when transformed with an expression plasmid harboring the pilin gene (pilE1) of Neisseria gonorrhoeae MS11, was able to express and assemble gonococcal pilin monomers into surface-associated pili, as judged by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, immunoblotting, and immunoelectron microscopy. Concomitant with the expression of gonococcal pili...

Many different species of the family Enterobacteriaceae have been shown to possess fimbrial appendages which emerge from the bacterial cell as straight, rigid organelles distinct from flagella (14). The role of fimbriae (synonym: pili) as adhesins in both enterotoxigenic and uropathogenic isolates of Escherichia coli has been demnonstrated, and thus the identification of such appendages as viru...
