Chemistry and pharmacology of benzodioxanes

Benzodioxane represents a series of synthetic and natural compounds of considerable medicinal importance. Compounds containing dioxane ring systems exhibited different biological activities such as antihepatotoxic, -adrenergic blocking agent, neuroleptic, anti-inflammatory, hypolipidemic and D 2 antagonist/5-HT 1A partial agonist activity. The literature concerning chemistry of benzodioxanes chemistry is scattered and has not been reviewed so far. The present article covers the various methods of preparing 1, 3-and 1, 4-benzodioxane ring system, different chemical reactions, and their different biological activities. 2008 Trade Science Inc. -INDIA

An efficient method for the preparation of various 2-substituted-1,4-benzodioxanes by CuBr-catalyzed tandem reactions of 2-((o-iodophenoxy)methyl)oxiranes with phenols has been developed. The reaction involves the ring-opening process of 2-((2-iodophenoxy)methyl)oxirane followed by an intramolecular C-O cross coupling cyclization.
