Growth Media for Typing Group a Streptococci.

چکیده
It is frequently not possible to type group A streptococei. Only 33% of group A streptococei from Nashville children in a recent study (Quinn and Martin, Am. J. Hyg. 73:193, 1961) were typable, and there have been years when as few as 5% could be typed. This study was designed to test one possible reason for failure to type certain strains of group A streptococci, namely, inadequacy of growth medium. Typable and nontypable strains of group A were studied, the former to compare the quantity of M protein produced in each medium and the latter to see whether they would become typable. Growth media were Todd-Hewitt Broth (BBL), 10 ml (TH); Todd-Hewitt Broth plus 2.2% sucrose, 10 ml (TH &S); Todd-Hewitt Broth, 40 ml (TH 40); and 0.5% yeast extract (Difco), 10 ml (Y). Each tube was inoculated with a 3-mm loopful of a broth culture of the strain being tested, and was incubated at 37 C for 18 hr. Acid extracts for serological grouping and typing were prepared according to the method of Swift et al. (J. Exptl. Med. 76:127, 1942). Extracts of typable strains were diluted 1:1, 1:2, 1:4, and 1:8 in 1 N saline before testing with their homologous antisera, to obtain an estimate of the quantity of M protein. Nontypable strains were tested with 34 types of antisera available from the Communicable Disease Center, At-

Rapid slide coagglutination test for identifying and typing group B streptococci.
A Cowan I strain of Staphylococcus aureus was labeled with either group B streptococcal grouping or typing antiserum. These antibody-labeled reagent cells (ARC) were used in a slide coagglutination test to identify and type group B streptococci from blood agar plates. All streptococci were also identified by the standard Lancefield capillary precipitin test. In a blind study, all 141 group B st...
متن کاملModified method for serological identification of group B streptococci.
Extracts from 10- and 40-ml cultures compared favorably for serological identification of group B streptococci. Typing by gel diffusion proved reliable and efficient. These modifications save time, media, and antisera.
متن کاملRecovering streptococci from the throat, a practical alternative to direct plating in remote tropical communities.
Throat swabs are regarded as the "gold standard" for diagnosing streptococcal pharyngitis and for surveillance research. Culturing throats in remote tropical settings is logistically difficult, and these settings are commonly burdened by high rates of streptococcal disease. The survival of streptococci on swabs may depend on whether they are of "throat" or "skin" type, as determined by emm patt...
متن کاملTyping Group a Hemolytic Streptococci
Two methods of typing group A hemolytic streptococci have been described: slide agglutination and anti-M precipitin tests. The first, developed by Griffith (1), is more extensively used. I t has the distinct advantages of requiring small amounts of serum and culture, and of a quick reaction time when all conditions are favorable. I t has certain disadvantages: The granular growth of some strain...
متن کاملPatterns of carriage of group B streptococci in genitourinary medicine clinic patients.
Anogenital carriage of group B streptococci was found in 46% (57/125) of men and 38% (68/179) of women attending a genitourinary medicine clinic. Colonisation with group B streptococci was more common in patients who had a history of more than one sexual partner in the preceding three months, but was not related to any previous sexually transmitted infection. Group B streptococci were isolated ...
