چکیده

Protein kinases (Rabinowitz, 1962) catalysing the transfer of the terminal phosphate of ATP to phosvitin, casein and possibly other phosphoproteins have been found in mammalian liver (Burnett & Kennedy, 1954), mammary gland (Sundararajan, Sampath-Kumar & Sarma, 1960), erythrocyte membranes (Judah, Ahmed & McLean, 1962) and brain (Rabinowitz & Lipmann, 1960). The following study of a protein kinase from cerebral cortex was prompted by a number of observations suggesting that a phosphoprotein may be involved in the transport of cations across the cell membrane. For example, on the application of electrical pulses to slices of cerebral cortex in vitro, the incorporation of added [32P]phosphate into a tissue phosphoprotein increases concomitantly with

Protein kinases in HeLa cells and in human cervix carcinoma.

Extracts of HeLa cell fractions were analyzed by DEAE- and phospho-cellulose chromatography for their range of cyclic AMP-dependent and -independent protein kinase activities phosphorylating histone and/or phosvitin; extractions were by phosphate buffered saline (soluble protein kinases) and the non-ionic detergent NP-40 (membrane-bound protein kinases). The soluble fraction contained (i) cycli...

Transcriptional effects of metal ions on the bovine oxytocin and the thymidine kinase-ERE promoter through the estrogen receptor a in MDA-MB 231 breast cancer cell line

BACKGROUND: Some of metal ions as environmental pollutants show estrogenic activity. This xenostrogenic compounds can be caused carcinogenicity in organs. The mechanism of carcinogenicity of metal ions is not clarified. OBJECTIVES: In this study, we investigated the Transcriptional effects of variety of metal ions on the bovine oxytocin and the thymidine kinase-ERE promoter by estrogen receptor...

Structural studies of phosvitin in solution and in the solid state.

Phosvitin, a highly phosphorylated glycoprotein, represents the major fraction of hen egg yolk phosphoproteins. Circular dichroism, Fourier transform infrared spectroscopy, and Fourier transform infrared photoacoustic and fluorescence spectroscopic methods were employed to determine the secondary structure of the protein in both the solid and solution phases. This was supplemented by a Chou-Fas...

Phosvitin kinase from the liver of the rooster. Purification and partial characterization.

Phosvitin kinase, the enzyme that phosphorylates the egg yolk protein phosvitin, has been purified more than SOOO-fold from the supernatant of rooster liver, a tissue that synthesizes phosvitin only after estrogen administration. The purified enzyme prepared from the estrogen-treated liver was recovered in high yield (49%) and was very stable at 04”. It sedimented in sucrose as a single peak wi...

Staining acidic phosphoproteins (phosvitin) in electrophoretic gels.

The principal phosphoglycoproteins of avian and amphibian egg yolk, known as phosvitins, have been examined extensively in developmental studies of estrogenic induction of protein synthesis (1) and, more recently, as substrates for protein kinases which phosphorylate the basic nuclear proteins (2,3). Phosvitin has few aromatic and basic amino acids (4) which can be detected by uv absorbance or ...
