چکیده

pyrimidine-specific nucleosidases which degraded radiola beled precursors used for measuring DNA and RNA synthe sis. Results of studies reported herein, and findings of others, demonstrate that CLC as well as peripheral blood leuko cytes of healthy subjects and patients with uncontrolled lymphocytic or myelocytic leukmeia release nucleosidases, and possibly other enzymes, when these cells are main tamed for a short time as a dense population. Failure to account for the possible presence of such enzymes in cul ture supernatants or cell homogenates may lead to the erroneous interpretation of data obtained in studies at tempting to characterize certain thermolabile lymphokines or chalones.

Elaboration of pyrimidine-specific nucleosidases by human lymphoblastoid cells of established cultures.

Pyrimidine-specific nucleosidases were released rapidly by human lymphoblastoid cells of established cultures when incubated under certain culture conditions having no adverse affect on their viability or morphology. Nucleosidase production was not restricted to any particular type of lymphoblastoid line; enzymes with a high level of activity were elaborated by cells of cultures initiated from ...

Lymphoblastoid cell lines: a continuous in vitro source of cells to study carcinogen sensitivity and DNA repair

Obtaining a continuous source of normal cells or DNA from a single individual has always been a rate limiting step in biomedical research. Availability of Lymphoblastoid cell lines (LCLs) as a surrogate for isolated or cryopreserved peripheral blood lymphocytes has substantially accelerated the process of biological investigations. LCLs can be established by in vitro infection of resting B cell...

Supportive Effects of Human Embryonic Fibroblast Cell Lines on Growth and Proliferation of EBV-Transformed Lymphoblastoid Cells

Human diploid fibroblast cells produce a spectrum of necessary growth factors and extracellular matrix (ECM) components essential for growth and proliferation of a variety of other cell types. In this study, the effect of five human embryonic fibroblast cell lines, isolated from liver, lung, skin and foreskin tissues, was investigated. A coculture system analyse was employed to cloning efficien...

Specificity of Mouse Uridine Phosphorylase*

Two distinct pyrimidine nucleosidases, isolated from Escheri&a coli, show complete specificity for the pentose moiety of the nucleoside. Pyrimidine deoxyribonucleoside phosphorylase (1) shows complete specificity for the deoxyribosyl group, and uridine phosphorylase (2) for the ribosyl group. Mammalian uridine phosphorylase preparations (3, 4), however, have appreciable deoxyuridine phosphoryla...

Molecular Docking Based on Virtual Screening, Molecular Dynamics and Atoms in Molecules Studies to Identify the Potential Human Epidermal Receptor 2 Intracellular Domain Inhibitors

Human epidermal growth factor receptor 2 (HER2) is a member of the epidermal growth factor receptor family having tyrosine kinase activity. Overexpression of HER2 usually causes malignant transformation of cells and is responsible for the breast cancer. In this work, the virtual screening, molecular docking, quantum mechanics and molecular dynamics methods were employed to study protein–ligand ...
