چکیده

Five different isolates of Pseudomonas aeruginosa were obtained from surgical specimens and minced meat. The isolates were tested for the production of the blue pigment; pyocyanin. Considerable amounts of blue pigment were produced by P. aeruginosa isolates when grown on the four tested media. Müller-Hinton agar was further used for growth, pigment production and sensitivity tests. Pigment production began during the first 24 hrs of growth and maximal pigment production was achieved after 48 hrs by isolates No. 1, 2 and 3. At this time, isolate No. 5 produced only moderate level of pyocyanin. While isolate No. 4 reached the highest yield after 72 hrs. Pyocyanin pigments obtained from culture supernatants of the tested P.aeruginosa isolates “grown in peptone water liquid medium” by serial chloroform extractions reached about 62.8μg/ml. The growth of all Gram-positive bacteria and Candida species was completely inhibited when cultivated on the agar plates containing the blue pigment. Whereas, Gram negative bacteria; Klebsiella pneumoniae and P. aeruginosa were resistant to pyocyanin. Salmonella typhi and Proteus mirabilis were intermediately affected. Thus, the five tested P. aeruginosa isolates were resistant to their own produced pyocyanin pigments. In a Conclusion, the Gram-positive bacteria were more susceptible as a group to the antibiotic action of pyocyanin than were the Gram negative bacteria. The antibacterial and antifungal nature of the pigment is attractive for the topical treatment of wound infections.

Drug resistance of isolated strains of Pseudomonas Aeruginosa from burn wound infections to selected antibiotics and disinfectants

Background and Objectives: Infection is the most common problem following burn injury. Selection and dissemination of intrinsic and acquired resistance mechanisms increase the probability of burn wound colonization by resistant species including Pseudomonas aeruginosa. Multi-drug resistant Pseudomonas ae...

Inhibitory Effect of Iron Oxide Nanoparticles on CTX-M Gene Expression in Extended-Spectrum βeta-Lactamase-Producing Pseudomonas aeruginosa Isolated from Burn Patients

Background and Objective: Burn wound infections caused by Pseudomonas aeruginosa exhibiting β-lactam antibiotic resistance are one of the greatest challenges of antimicrobial treatment. In this context, P. aeruginosa strains harboring resistance mechanisms, such as production of extended-spectrum beta-lactamases have the highest clinical impact no the management of burn wound infections. The ai...

Effect of fetal and adult bovine serum on pyocyanin production in Pseudomonas aeruginosa isolated from clinical and soil samples

Objective(s): Pyocyanin is a blue-greenish redox-active pigment, produced by Pseudomonas aeruginosa, with a wide range of biological and biotechnological applications. Pyocyanin biosynthesis is regulated by the quorum-sensing (QS) system in which the expression of QS genes and QS-controlled virulence genes may be affected by serum as a complex medium. In the current study, effects of adult bovi...

Determination antimicrobial resistance profile of Pseudomonas aeruginosa strains isolated from hospitalized patients in Taleghani Hospital (Ahvaz, Iran) from 2011-2012

Background & Objective: Pseudomonas aeruginosa is a rod-shaped, Gram-negative, glucose-nonfermenting aerobic bacterium. It is widespread in natural environments and it is an opportunistic pathogen for humans that can lead to a broad spectrum of disease such as urinary, burn, respiratory infections, and septicemia. The aim of this study was to determe antibiotic resistance profile of P. aerugino...

Pyocyanin production by Pseudomonas aeruginosa confers resistance to ionic silver.

Silver in its ionic form (Ag+), but not the bulk metal (Ag0), is toxic to microbial life forms and has been used for many years in the treatment of wound infections. The prevalence of bacterial resistance to silver is considered low due to the nonspecific nature of its toxicity. However, the recent increased use of silver as an antimicrobial agent for medical, consumer, and industrial products ...
