چکیده

The mechanism by which the mammalian kidney excretes inorganic phosphate has not been fully elucidated. The only micropuncture data available are those of Walker and Hudson (1), and they deal with the mode of phosphate excretion in two amphibian species. These investigators reported that the inorganic phosphate concentration rises progressively along the proximal and distal nephron of the frog and Necturus. In the frog this rise can be accounted for by tubular fluid reabsorption, but in the Necturus the secretion of phosphate could not always be excluded. Evidence obtained by clearance experiments in vertebrates indicates significant species differences. Thus, renal tubular phosphate secretion has been conclusively demonstrated in certain fish (2-4), the alligator (5), and the chicken (6). The mechanism of phosphate excretion in the mammalian kidney, on the other hand, has remained, to some extent, controversial. According to Pitts and Alexander, in the dog (7) there is a maximal tubular reabsorptive capacity for phosphate, the renal transfer of phosphate being achieved by a combination of glomerular filtration and subse-

Renal phosphate adaptation in uraemic dogs with a remnant kidney.

1. Clearance and micropuncture studies were performed in 27 dogs made uraemic by segmental infarction to examine the factors responsible for phosphate adaptation in chronic renal failure. 2. The animals were studied before and after extracellular volume expansion to 10% of body weight in the presence and absence of parathyroid glands. The results were compared with 19 normal dogs studied under ...

The effects of glucose and insulin on renal electrolyte transport.

The effects of hyperglycemia and hyperinsulinemia on renal handling of sodium, calcium, and phosphate were studied in dogs employing the recollection micropuncture technique. Subthreshold sustained hyperglycemia resulted in an isonatric inhibition of proximal tubular sodium, fluid, calcium, and phosphate reabsorption by 8-14%. Fractional excretion of sodium and phosphate, however, fell (P is le...

Effect of phlorhizin on renal glucose and phosphate transport in the dog.

1. Clearance and micropuncture studies were performed in 19 thyroparathyroidectomized dogs to examine the inter-relationship between the renal transport of sodium, glucose and phosphate. 2. All experiments were carried out before and after the intravenous administration of phlorhizin [7 mg (15 pnol)/kgl with a sustaining infusion of the same dose/h. Thirteen dogs were studied during hydropenia ...

Evidence for an intrinsic renal tubular defect in mice with genetic hypophosphatemic rickets.

To investigate the role of parathyroid hormone (PTH) and(or) an intrinsic renal tubular reabsorptive defect for phosphate in mice with hereditary hypophosphatemic rickets, we performed clearance and micropuncture studies in hypophosphatemic mutants and nonaffected littermate controls. Increased fractional excretion of phosphate in mutants (47.2+/-4 vs. 30.8+/-2% in controls) was associated with...

Direct micropuncture evidence that matrix extracellular phosphoglycoprotein inhibits proximal tubular phosphate reabsorption.

BACKGROUND Matrix extracellular phosphoglycoprotein (MEPE) is a putative phosphatonin that we have shown in previous studies to be phosphaturic in rats. Its site of action in the nephron remains to be confirmed. METHODS We made micropuncture collections from late proximal convoluted tubules in anaesthetized rats to assess directly the effect of MEPE on phosphate reabsorption in the proximal t...
