A Micro Method for Serum Cholinesterase.
نویسندگان
چکیده
A simple colorimetric method for determining serum or plasma cholinesterase has been described. The cholinesterase acts on acetyithiocholine to release thiocholine, which reacts with 5:5-dithiobis(2-nitrobenzoic acid) to produce a yellow color. The activity is measured after incubation for 3 mm. at 370; the reaction is stopped by the excess addition of quinidine sulfate. The difference in absorbance between the blank and test sample is then measured at 412 mL. Duplicate determinations can be run on 0.1 ml. of serum or plasma, and the method is well suited to laboratories using routine procedures.

Rapid Estimation of Serum Cholinesterase Activity Using the Astrup Micro Equipment.
A rapid micro technique for the estimation of serum cholinesterase is described. Acetylcholine bromide is incubated with serum within the capillary of the Astrup electrode. The enzyme hydrolyses the substrate with the liberation of acetic acid. This causes a fall of pH which is seen on the galvanometer of the instrument and the rate of this fall is shown to be proportional to enzyme concentrati...
متن کاملSerum Paraoxonase 1 Activity in Patients with Organophosphate Poisoning: A Potential Indicator of Prognosis
Background: Human serum paraoxonase 1 (PON1) hydrolyzes organophosphate (OP) compounds and so significantly alters an individual’s susceptibility to the toxicity of these chemicals. The study was designed to assess the serum PON1 activity in a series of patients with OP poisoning. Methods: Suspected OP poisoning patients presented within 6 hours of...
متن کاملComparative Measurement of Serum Acetyl Cholinesterase Enzyme Using Three Different Methods
Introduction: IAChE (acetylcholinesterase) is one of the plasma’s enzymes that plays a crucial role in nervous signal transduction in synapses and is responsible for the hydrolysis of Ach. Material & Method: We investigated three methods for the determination of AChE in human blood serum, using three different assays. Michel methods are based on the measurement of hydrogen ion produced in pro...

Introduction & Objective: Diazinon (DZN) is an organophosphate insecticide that one of the mechanisms of toxicity is the inhibition of cholinesterase. The aim of the present study was to evaluate the effects of diazinon on cholinesterase activity in blood serum and erythrocytes of male and female rats and to assess the protective role of vitamin E. Materials & Methods: In this experimental stud...
متن کاملChanges of serum level of acetylcholinesterase enzyme in lipopolysaccharide-induced model of depression in mice
Background and Objective: Depression is a common disorder, especially in developed countries. Functional changes of cholinesterase are involved in pathogenesis of some brain disorders. Until now, exact association of these changes with depression has not been determined. This study was conducted to evaluate the changes of serum cholinesterase in lipopolysaccharide (LPS)-induced model of depress...
