چکیده
WSA2 from mycobacteria have recently been shown to have potent biological activity ( I , 2, 5). These adjuvants are nontoxic in vitro and may prove useful in amplification of the immune response. Adam et a!. ( 1) reported that water-soluble fractions that were extracted from the myco bacterial cell walls were nontoxic and were capable of increasing circulating antibodies and of inducing delayed hypersensitivity to ovalbumin ( 1). WSA have also been shown to increase the immune response to sheep red cells, 2,4-dinitrophenol-dextran, and 2,4-dinitrophenol-edestin (7) and to viral antigens (2). Evidence for the potentiation of lymphocyte transformation and in vitro immunization by WSA is provided here.

IN VITRO STUDY OF AN ENDOGENOUS IMMUNOSUPPRESSOR FACTOR DERIVED FROM HUMAN OR BOVINE SERUM
The effects of the human and bovine LSF (derived from sera) as well as their purified fractions were studied on murine lymphocytes reactions indicated by blast transformation (BT) assay, mixed lymphocyte culture (MLC) and IgG synthesis. The results indicated that bovine lipid suppressor factor (LSF) has significant immunosuppressive activity on lymphocytes proliferation both in BT and MLC ...
متن کاملLymphocyte transformation test in leprosy: decreased lymphocyte reactivity to Mycobacterium leprae in lepromatous leprosy, with no evidence for a generalized impairment.
Untreated leprosy patients were examined with respect to lymphocyte transformation in vitro after stimulation with mycobacterial and other microbial antigens, allogeneic lymphocytes, or nonspecific mitogens. Methods were used to circumvent technical variability. The results were compared with those obtained in controls matched for age, sex, race, and environment. No evidence was found for a gen...
متن کاملAcanthamoeba polyphaga-Enhanced Growth of Mycobacterium smegmatis
BACKGROUND Mycobacterium smegmatis is a rapidly-growing mycobacterium causing rare opportunistic infections in human patients. It is present in soil and water environments where free-living amoeba also reside, but data regarding M. smegmatis-amoeba relationships have been contradictory from mycobacteria destruction to mycobacteria survival. METHODOLOGY/PRINCIPAL FINDINGS Using optic and elect...
متن کاملGenetic alteration of Mycobacterium smegmatis to improve mycobacterium-mediated transfer of plasmid DNA into mammalian cells and DNA immunization.
Mycobacteria target and persist within phagocytic monocytes and are strong adjuvants, making them attractive candidate vectors for DNA vaccines. We characterized the ability of mycobacteria to deliver transgenes to mammalian cells and the effects of various bacterial chromosomal mutations on the efficiency of transfer in vivo and in vitro. First, we observed green fluorescent protein expression...

Abstract Background and purpose: Different laboratories have considered different maturation factors for producing human dendritic cells (DC) from peripheral blood monocytes. In this study, we comprehensively investigated the effect of adding poly (I:C) to standard maturation stimulus, i.e. MCM and TNF-α on the induction of T cell response. Materials and methods: Peripheral blood monocytes ...
