چکیده

UNLABELLED PREMISE OF THE STUDY Microsatellite primers were developed in the endangered tree species Taxus wallichiana from Nepal to investigate regional genetic differentiation, local genetic diversity, and gene flow for the conservation of this species under climate- and land-use change scenarios in mountain regions of Nepal. • METHODS AND RESULTS We developed 10 highly polymorphic microsatellite markers from 454 DNA sequencing. Characterization of the new microsatellite loci was done in 99 individuals collected from three valleys with different climatic regimes. The number of alleles per locus varied from four to 12. Observed heterozygosity of populations, averaged across loci, ranged from 0.30 to 0.59. • CONCLUSIONS The new markers provided by this study will substantially increase the resolution for detailed studies in phylogeography, population genetics, and parentage analysis.

Development of microsatellite loci for Taxus wallichiana var. wallichiana (Taxaceae) and cross-amplification in Taxaceae.

Nine polymorphic microsatellite loci were isolated and characterized in Taxus wallichiana var. wallichiana, an endangered species in China. The number of alleles per locus ranged from 2 to 20. Observed and expected heterozygosities varied from 0.0260 to 0.5325 and 0.3603 to 0.9231, respectively. Positive cross-amplification of the 9 loci was observed in 2 other varieties of T. wallichiana and 4...

Microsatellite loci for an old rare species, Pseudotaxus chienii, and transferability in Taxus wallichiana var. mairei (Taxaceae)1

UNLABELLED PREMISE OF THE STUDY Microsatellite loci were developed for Pseudotaxus chienii, an old rare species endemic to China, and which provided a useful tool for investigating the patterns of population genetic structure, phylogeography, evolutionary history, and adaptive potential. Transferability was assayed in the related species, Taxus wallichiana var. mairei. • METHODS AND RESULT...

Development of polymorphic microsatellite markers for Japanese yew, Taxus cuspidata, and T. cuspidata var. nana (Taxaceae)1

PREMISE OF THE STUDY Taxus cuspidata (Taxaceae), which is well known for the effective anticancer metabolite paclitaxel (e.g., taxol), is an evergreen needle-leaved tree widely distributed in eastern Eurasia including Japan. We developed 15 microsatellite markers from this species and confirmed their utility for the dwarf variety nana, which is common in alpine regions along the Sea of Japan. ...

Isolation and Characterization of Microsatellite Markers from Endangered Species (Camelus bactrianus)

Iranian bactrian camel (Camelus bactrianus) is an endangered livestock breed with distribution in northwest of Iran. Microsatellites are a powerful marker for animal genetic and cell line identification and population genetic study. In this study, after producing more than 40 Camelus bactrianus fibroblast cell lines, microsatellites loci from the genome of Iranian Camelus bactrianus cell lines ...

Isolation and Characterization of Microsatellite Markers in the Endangered Species Taxus wallichiana Using the FIASCO Method

The Himalayan yew, Taxus wallichiana Zucc., is an endangered species with a scatted distribution in the Eastern Himalayas and southwestern China. In the present study, 10 microsatellite markers from the genome of T. wallichiana were developed using the protocol of fast isolation by amplified fragment length polymorphism of sequences containing repeats (FIASCO). Polymorphism of each locus was as...
