Fluoride inhibition of the initiation of protein synthesis in the reticulocyte lysate cell-free system.

چکیده
KF (30 mM) strongly inhibits polypeptide chain initiation in the reticulocyte lysate cell-free system.. Chain elongation is partially inhibited but proceeds to a significant extent with little initiation of new chains. Polysome breakdown is incomplete after incubations as long as 20 min. Under these conditions deacylated tRNA-Met accumulates in a fraction sedimenting faster than 120 S and thus may be associated with ribosomes bound to mRNA. Incubation of the system with KF results in the accumulation of a complex which can initiate synthesis of polypeptide chains in the presence of aurintricarboxylate; KF thus inhibits a step in initiation after that inhibited by aurintricarboxylate. The accumulation of deacylated tRNA-Met is correlated with the accumulation of the aurintricarboxylate-resistant complex. Both phenomema are dependent on KF and both are inhibited by aurintricarboxylate in the same range of concentrations which inhibit initiation of protein synthesis.

Inhibition of binding to initiation complexes of nascent reovirus mRNA by double-stranded RNA-dependent protein kinase.
A coupled, cell-free system for the transcription and translation of reovirus mRNA was developed. Activated reovirions were incubated with reticulocyte lysate and an appropriate energy mix. Active transcription was obtained, but protein synthesis was inhibited after a short lag even by low concentrations of reovirions. This inhibition was abolished by the addition of the kinase inhibitor 2-amin...
متن کاملCharacterization of cell-free protein-synthesis systems from undeveloped and developing Artemia embryos.
We have developed and characterized translationally active cell-free systems from Artemia embryos at different developmental times. The optimized lysates from 16 h-developed embryos incorporated radiolabelled amino acids into polypeptides for up to 120 min. The polypeptides synthesized ranged in Mr from 150,000 to 10,000, suggesting that the endogenous mRNA was capable of directing the synthesi...
متن کاملInhibitory effect of alpha-fetoprotein on protein synthesis in a reticulocyte lysate cell-free system.
The inhibitory effect of human alpha-fetoprotein on the protein synthesis (incorporation of [3H]leucine into the total protein fraction) weas observed in a rabbit reticulocyte lysate cell-free system. The incorporation was inhibited to 50% of the control level by 14-16 microM alpha-fetoprotein, and decreased to about 15% at a concentration higher than 40 microM. An almost identical dose depende...
متن کاملStructure and function of initiation complexes which accumulate during inhibition of protein synthesis by fluoride ion.
Incubation of the reticulocyte lysate cell-free system with KF results in the accumulation in polysomes of complexes containing deacylated tRNAMet and of complexes which can initiate globin chains in the presence of aurintricarboxylate. Degradation of these polysomes with T1RNase yields both 40 S and 80 S particles, and tRNAMet is found in both of these fractions. When the 80 S particles are re...
متن کاملChloroquine inhibits heme-dependent protein synthesis in Plasmodium falciparum.
A cell-free protein-synthesizing system has been reconstituted using the S-30 fraction or ribosomes and the S-100 fraction from Plasmodium falciparum. Addition of heme in vitro stimulates cell-free protein synthesis strikingly. Chloroquine inhibits the heme-dependent protein synthesis in the parasite lysate. The drug has also been found to inhibit parasite protein synthesis in situ at therapeut...
