New measurements of DNA twist elasticity.

چکیده
The symmetries of the DNA double helix require a new term in its linear response to stress: the coupling between twist and stretch. Recent experiments with torsionally constrained single molecules give the first direct measurement of this new material parameter. We extract its value from a recent experiment. Finally, we sketch the effect of constrained twist on entropic elasticity of DNA arising from the connection between Link, Twist, and Writhe.

Quantifying how DNA stretches, melts and changes twist under tension
In cells, DNA is constantly twisted, bent and stretched by numerous proteins mediating genome transactions. Understanding these essential biological processes requires in-depth knowledge of how DNA complies to mechanical stress. Two important physical features of DNA, helical structure and sequence, are not incorporated in current descriptions of DNA elasticity. Here we connect well-defined for...

The symmetries of the DNA double helix require a new term in its linear response to stress: the coupling between twist and stretch. Recent experiments with torsionally-constrained single molecules give the first direct measurement of this important material parameter. We extract its value from a recent experiment of Strick, et al. and find rough agreement with an independent experimental estima...
متن کاملElasticity Theory of a Twisted Stack of Plates
We present an elastic model of B-form DNA as a stack of thin, rigid plates or base pairs that are not permitted to deform. The symmetry of DNA and the constraint of plate rigidity limit the number of bulk elastic constants contributing to a macroscopic elasticity theory of DNA to four. We derive an effective twist-stretch energy in terms of the macroscopic stretch ǫ along and relative excess tw...
متن کاملDNA Twist Stability Changes with Magnesium(2+) Concentration.
To understand DNA elasticity at high forces (F>30 pN), its helical nature must be taken into account, as a coupling between twist and stretch. The prevailing model, the wormlike chain, was previously extended to include this twist-stretch coupling. Motivated by DNA's charged nature, and the known effects of ionic charges on its elasticity, we set out to systematically measure the impact of buf...
متن کاملBending and Twisting Elasticity of DNA
We derive the elastic theory suitable for describing the free energy required to deform a stiff helical molecule with the symmetry of DNA. At quadratic levels in the strains we find, in addition to the independent bending and torsional energies incorporated in previous theories, a previously unknown coupling between twist and bend. If the backbone is given constant curvature, minimization of th...
