A trypsin inhibitor cDNA from a novel source, snail medic (Medicago scutellata L.): cloning and functional expression in response to wounding, herbivore, jasmonic and salicylic acid

چکیده
A cDNA clone coding for the trypsin inhibitor MsTI from snail medic (Medicago scutellata L.) was isolated by RT-PCR using poly(A)+ mRNA purified from immature seeds. A partial cDNA fragment was produced using degenerated oligonucleotides designed from the available MsTI amino acid sequence. The full-length cDNA was obtained by RACE-PCR experiments. A putative signal peptide for subcellular localization is part of the open reading frame. This is the first example of a cloned cDNA sequence from the pasture legume M. scutellata. Southern analysis revealed the presence of a small size multigene family. When polyclonal antibodies raised against the purified seed protein were utilized in Western blot analyses, a 6.9 kDa protein from crude extract was recognized. Expression studies were carried out in order to define the role of the MsTI inhibitor in the natural defense system developed by plants against pathogen attacks. A constitutive expression pattern of the MsTI gene was observed in stems, while the transcript was absent in both root and leaf tissues. Local and systemic accumulation of MsTI mRNA was detected in leaves in response to mechanical wounding and exposure to Hypera postica larvae. The insect damage determined a faster response compared to mechanical treatment. In addition, induction of the MsTI gene expression after exogenous applications of salicylic acid and jasmonic acid was observed. An insect bioassay, performed with larvae of the insect pest Hyphantria cunea fed with the purified protein, was carried out. Results from this study suggest a role of MsTI protein in plant defense reactions, highlighting its possible use for biotechnological applications. © 2004 Elsevier Ireland Ltd. All rights reserved.

Seed Proteinase Inhibitors from Annual Medics Active against Insect Pests
11-19 ABSTRACT Several different species of annual medics have been tested for the occurrence and concentration of trypsin inhibitors in the seed. Trypsin Inhibitory Activity (TIA) has been in fact related to the level of resistance to biotic and abiotic stresses. Wide variation was found for TIA among species, with the highest values shown by the snail medic (Medicago scutellata). After a simp...

BACKGROUND Snail medic (Medicago scutellata L.) seeds exhibit a significantly higher content of a trypsin inhibitor than other Medicago species. This inhibitor belongs to the Bowman-Birk family of serine protease inhibitors (BBI) and exhibits a good sequence homology with the BBI from soybean, while presenting some differences. It has been suggested that BBIs have antitumoral and radio-protecti...
متن کاملThe effect of salicylic acid and jasmonic acid on the symptoms of the disease and the expression of defense genes on a greenhouse cucumber infected with CMV
Cucumber is one of the most important vegetable and summer crops economically and is the fourth most cultivated plant in the world after tomatoes and potatoes. In this study, the effect of plant inducers including salicylic acid and jasmonic acid on the viral disease of greenhouse cucumber with Cucumber mosaic virus (CMV) and also the expression of defense genes was investigated. Treatments inc...

Gel electrophoretic analysis of the proteinase inhibitor proteins induced in tomato leaves by airborne methyl jasmonate (EE Farmer, CA Ryan [1990] Proc Natl Acad Sci USA 87: 7713-7716) revealed the new appearance of inhibitors I and II and two other, higher molecular mass proteins (63.5 and 87 kilodaltons). Northern analysis of methyl jasmonate-induced inhibitors I and II mRNAs in tomato (Lycop...

In a wild tobacco plant, Nicotiana attenuata, two mitogen-activated protein kinases (MAPKs), salicylic acid-induced protein kinase (SIPK) and wound-induced protein kinase (WIPK), play central roles in modulating herbivory-induced phytohormone and anti-herbivore secondary metabolites. However, the identities of their upstream MAPK kinases (MAPKKs) were elusive. Ectopic overexpression studies in ...
