T-type calcium channels regulate cortical plasticity in-vivo NR-D-08-7049

چکیده
T-type voltage-dependent calcium channels may play an important role in synaptic plasticity, but lack of specific antagonists has hampered investigation into this possible function. We investigated the role of the T-type channel in a canonical model of in-vivo cortical plasticity triggered by monocular deprivation. We identified a compound (TTA-I1) with subnanomolar potency in standard voltage clamp assays and high selectivity for the T-type channel. When infused intracortically, TTA-I1 reduced cortical plasticity triggered by monocular deprivation while preserving normal visual response properties. These results show that the T-type calcium channel plays a central role in cortical plasticity. NeuroReport 20:257–262 c 2009 Wolters Kluwer Health | Lippincott Williams & Wilkins. Involvement of voltage-dependent calcium channels in synaptic plasticity of the rat visual cortex
In this study, involvement ofvoltage-dependent calcium channels in LTP of responses of rat visual cortex slices was analyzed. Field potentials including EPSP1 and EPSP2 from layers II/III were recorded through stimulation of layer IV. Whereas nifedipine, a L-type calcium channel blocker (L-VDCC), did not considerably affect the LTP of responses, but Ni2+, a relatively selective blocker of T-typ...

متن کاملAstrocyte calcium signaling transforms cholinergic modulation to cortical plasticity in vivo.
Global brain state dynamics regulate plasticity in local cortical circuits, but the underlying cellular and molecular mechanisms are unclear. Here, we demonstrate that astrocyte Ca(2+) signaling provides a critical bridge between cholinergic activation, associated with attention and vigilance states, and somatosensory plasticity in mouse barrel cortex in vivo. We investigated first whether a co...

Evidence suggests that during bursts of action potentia...
متن کاملSynaptic strength at the temporoammonic input to the hippocampal CA1 region in vivo is regulated by NMDA receptors, metabotropic glutamate receptors and voltage-gated calcium channels
The hippocampal CA1 region receives cortical information via two main inputs: directly via the perforant (temporoammonic) path (pp-CA1 synapse) and indirectly via the tri-synaptic pathway. Although synaptic plasticity has been reported at the pp-CA1 synapse of freely behaving animals, the mechanisms underlying this phenomenon have not been investigated. Here, we explored whether long-term poten...
