چکیده

Throughout all domains of life, nucleic acid-based epigenetic modification of DNA is established as a regulatory mechanism that can affect different processes ranging from the replicative mismatch repair to the transcriptional activity of genes. Modifications can be preserved during cell divisions and play important roles in development-with consequences of aberrant regulation of modification such as the promotion of diseases including cancerY-3J Several types of modification that are associated with varying biological roles, depending on the organism and cell type are known. Methylation of cytosine nucleobases at position 5 (mC, Figure 1 A) is the best-studied epigenetic mark in eukar-

Strategies and Clinical Applications of Next Generation Sequencing

Abstract DNA sequencing is one of the great valuable techniques in molecular biology, which can be used to detect the sequence of nucleotides in a DNA fragment. The high-throughput sequencing known as Next Generation Sequencing (NGS) revolutionized genomic research and molecular biology; therefore, the whole human genome can be sequenced with a low cost in several days. NGS technology is simi...

White Paper - Detecting DNA Base Modifications Using SMRT Sequencing

Traditionally, it has been a challenge to study the wide variety of base modifications that are seen in nature. Most high-throughput techniques focus only on cytosine methylation and involve both bisulfite sequencing to convert unmethylated cytosine nucleotides to uracil nucleotides, and comparison of sequence reads from bisulfite-treated and untreated samples. SMRT sequencing, in contrast, doe...

Introduction into the analysis of high-throughput-sequencing based epigenome data

Sequencing-based approaches now allow high-resolution, genome-scale investigation of cellular epigenetic landscapes. For example, mapping of open chromatin regions, post-translational histone modifications and DNA methylation across a whole genome is now feasible, and new non-coding regulatory RNAs can be sensitively identified via RNA sequencing. The resulting large-scale data sets promise to ...

Single molecule-level detection and long read-based phasing of epigenetic variations in bacterial methylomes

Beyond its role in host defense, bacterial DNA methylation also plays important roles in the regulation of gene expression, virulence and antibiotic resistance. Bacterial cells in a clonal population can generate epigenetic heterogeneity to increase population-level phenotypic plasticity. Single molecule, real-time (SMRT) sequencing enables the detection of N6-methyladenine and N4-methylcytosin...
