CD 1 d - mediated Recognition of an a - Galactosylceramide by Natural Killer T Cells Is Highly Conserved through Mammalian Evolution

چکیده
Natural killer (NK) T cells are a lymphocyte subset with a distinct surface phenotype, an invariant T cell receptor (TCR), and reactivity to CD1. Here we show that mouse NK T cells can recognize human CD1d as well as mouse CD1, and human NK T cells also recognize both CD1 homologues. The unprecedented degree of conservation of this T cell recognition system suggests that it is fundamentally important. Mouse or human CD1 molecules can present the glycolipid a -galactosylceramide ( a -GalCer) to NK T cells from either species. Human T cells, preselected for invariant V a 24 TCR expression, uniformly recognize a -GalCer presented by either human CD1d or mouse CD1. In addition, culture of human peripheral blood cells with a -GalCer led to the dramatic expansion of NK T cells with an invariant (V a 24 1 ) TCR and the release of large amounts of cytokines. Because invariant V a 14 1 and V a 24 1 NK T cells have been implicated both in the control of autoimmune disease and the response to tumors, our data suggest that a -GalCer could be a useful agent for modulating human immune responses by activation of the highly conserved NK T cell subset.

Valpha24-Jalpha18 invariant natural killer T cells (iNKT) are an evolutionary conserved sub-lineage of T cells with effector-memory phenotype that often express an NK cell surface antigen CD161 and is characterized by reactivity to self-glycolipids and alpha-galactosylceramide (alphaGalCer) that are presented by monomorphic HLA class-I-like molecule CD1d. Upon antigen recognition, iNKT cells ca...

We have recently identified alpha-galactosylceramide (alpha-GalCer) as a specific ligand for an invariant Valpha14/Vbeta8.2 T cell receptor exclusively expressed on the majority of Valpha14 NKT cells, a novel subset of lymphocytes. Here, we report that alpha-GalCer selectively activates Valpha14 NKT cells resulting in prevention of tumor metastasis. The effector mechanisms of the ligand-activat...

Natural killer (NK) T cells recognize lipid antigens in the context of the major histocompatibility complex (MHC) class 1-like molecule CD1 and rapidly secrete large amounts of the cytokines interferon (IFN)-gamma and interleukin (IL)-4 upon T cell receptor (TCR) engagement. We have asked whether NK T cell activation influences adaptive T cell responses to myelin antigens and their ability to c...

CD1d-restricted natural killer T (NKT) cells include two major subgroups. The most widely studied are Vα14Jα18(+) invariant NKT (iNKT) cells that recognize the prototypical α-galactosylceramide antigen, whereas the other major group uses diverse T-cell receptor (TCR) α-and β-chains, does not recognize α-galactosylceramide, and is referred to as diverse NKT (dNKT) cells. dNKT cells play importan...
