چکیده
Hemolysin BL (HBL) is the multi-component enterotoxin causing diarrheal type of Bacillus cereus food poisoning. The hbl operon of B. cereus was divided into 4 coding sequences (CDS) and identified as hblC, hblD, hblA and hblB genes consisting of 1,319, 1,221, 1,128 and 1,401 nucleotides encoding proteins of 439, 406, 375 and 466 amino acids with predicted sizes of 49.3, 43.8, 41.7 and 52.3 kDa, respectively. One hundred of B. cereus isolates were detected for the presence of each hbl gene by multiplex PCR and HBL production by discontinuous hemolysis on HBL sheep blood agar and Bacillus cereus Enterotoxin-Reverse Passive Latex Agglutination (BCET-RPLA) test kit. The results showed that 47 of 100 B. cereus isolates (47%) were detected by multiplex PCR with all hbl genes and gave positive result with RPLA test. Forty-one of 47 isolates (87.2%) produced discontinuous hemolysis on HBL sheep blood agar. However, this pattern was not observed clearly in 6 of 47 isolates (12.8%). In addition, B. cereus isolate E82/44 gave multiplex PCR positive with only 3 hbl genes, hblA, hblC and hblD, also showed positive result with RPLA test and produced discontinuous hemolysis. Keyword: Bacillus cereus, Hemolysin BL, Enterotoxin, Food poisoning.

Identification of hemolysin BL-producing Bacillus cereus isolates by a discontinuous hemolytic pattern in blood agar.
Bacillus cereus causes distinct exotoxin-mediated diarrheal and emetic food poisoning syndromes and a variety of nongastrointestinal infections. Evidence is accumulating that hemolysin BL is a major B. cereus virulence factor. We describe two methods for detection of hemolysin BL in crude samples and on primary culture media. In the first method, the highly unusual discontinuous hemolysis patte...
متن کاملDetection of enterotoxic Bacillus cereus producing hemolytic and non hemolytic enterotoxins by PCR test.
Nine strains belonging to Bacillus cereus group has been isolated from food and environmental samples. Their taxonomic position was confirmed by RFLP analysis of 16S rRNA gene digested with TaqI. The detection of DNA sequences encoding the hemolysin BL complex and enterotoxin NHE, was studied in Bacillus sp. isolates. Set of primers was used to amplify fragment of hblD gene by PCR. For the dete...

Bacillus cereusis an ubiquitous, spore-forming bacteria that can survive pasteurization and the majority of the heating processes used in the dairy industry. Besides, it is a pathogen responsible for different types of food poisoning. One type of foodborne disease caused by B.cereusis the diarrheal syndrome, which is caused by the ingestion of vegetative cells producing toxins in the small inte...
متن کاملSwarming behavior of and hemolysin BL secretion by Bacillus cereus.
An association between swarming and hemolysin BL secretion was observed in a collection of 42 Bacillus cereus isolates (P=0.029). The highest levels of toxin were detected in swarmers along with swarm cell differentiation (P=0.021), suggesting that swarming B. cereus strains may have a higher virulence potential than nonswarming strains.

Forty-seven strains representing 14 different Bacillus species isolated from clinical and food samples were grown in reconstituted infant milk formulae (IMF) and subsequently assessed for adherence to, invasion of, and cytotoxicity toward HEp-2 and Caco-2 cells. Cell-free supernatant fluids from 38 strains (81%) were shown to be cytotoxic, 43 strains (91%) adhered to the test cell lines, and 23...
