Bending and Twisting Elasticity of DNA

چکیده
We derive the elastic theory suitable for describing the free energy required to deform a stiff helical molecule with the symmetry of DNA. At quadratic levels in the strains we find, in addition to the independent bending and torsional energies incorporated in previous theories, a previously unknown coupling between twist and bend. If the backbone is given constant curvature, minimization of the free energy with respect to the twist degrees of freedom indicates that this coupling drives a decrease in the molecular twist, or an unwinding of the helix. New experiments are proposed to bring out the symmetry-breaking effecte of the twist-bend coupling: (i) ring closure experiments will indicate a helix repeat that becomes progressively more underwound for smaller rings, and (ii) gel mobilities of supercoiled rings of integral-helix-repeat length, with equal and opposite added linking numbers, will differ.

Bending and Twisting Elasticity: a Revised Marko-Siggia Model on DNA Chirality
A revised Marko-Siggia elastic model for DNA double helix [Macromolecules 27, 981 (1994)] is proposed, which includes the WLC bending energy and a new chiral twisting energy term. It is predicted that the mean helical repeat length (HRL) for short DNA rings increases with the decreasing of chain length; while for very long chains, their mean HRL is the same, independent of both the chain length...
متن کاملLocal elasticity of strained DNA studied by all-atom simulations.
Genomic DNA is constantly subjected to various mechanical stresses arising from its biological functions and cell packaging. If the local mechanical properties of DNA change under torsional and tensional stress, the activity of DNA-modifying proteins and transcription factors can be affected and regulated allosterically. To check this possibility, appropriate steady forces and torques were appl...
متن کاملOrigin of twist-bend coupling in actin filaments.
Actin filaments are semiflexible polymers that display large-scale conformational twisting and bending motions. Modulation of filament bending and twisting dynamics has been linked to regulatory actin-binding protein function, filament assembly and fragmentation, and overall cell motility. The relationship between actin filament bending and twisting dynamics has not been evaluated. The numerica...

The sequence–dependent elasticity of double-helical DNA on a nm length scale can be captured by the rigid base–pair model, whose strains are the relative position and orientation of adjacent base– pairs. Corresponding elastic potentials have been obtained from all–atom MD simulation and from high–resolution structural data. On the scale of a hundred nm, DNA is successfully described by a contin...

We have investigated the microsecond rotational dynamics of F-actin with transient phosphorescence anisotropy (TPA) spectroscopy, and analyzed the data to determine the relative contributions from rigid-body rotations and from intrafilament bending and twisting, using a theoretical model developed for DNA dynamics by Schurr and co-workers. The fits of the data to the model were constrained by i...
