An easily controlled method for staining mitochondria.

چکیده

THE great disadvantages of the standard staining methods for mitochondria are that they are very complicated and cannot be applied to different tissues until suitable times for the action of the various stains have been worked out by a long process of trial and error. The following method is easy to control and apply. Destaining and restaining are quickly carried out, times may be worked out on a single section, and, as each reagent is used for a single purpose (not, for example, as a simultaneous differentiator and stain), each stage may be controlled separately. Mitochondria are red, cytoplasm is pale blue or colourless, nuclei are medium blue, and plasmosomes (nucleoli) pink or red.

Fluorescent staining of fungal nuclei with a benzimidazol derivative.

A direct staining procedure is described for fluorescence microscopy of fungal nuclei, chromosomes and mitochondria. The fluorochrome is a benzimidazol derivative (33258 Hoechst) known to bind selectively to deoxyribonucleic acid at neutral pH. The advantages of 33258 Hoechst relative to Feulgen compounds used previously to stain these structures include a greater intensity of fluorescence, th...

Correlative Super-resolution and Electron Microscopy to Resolve Protein Localization in Zebrafish Retina

We present a method to investigate the subcellular protein localization in the larval zebrafish retina by combining super-resolution light microscopy and scanning electron microscopy. The sub-diffraction limit resolution capabilities of super-resolution light microscopes allow improving the accuracy of the correlated data. Briefly, 110 nanometer thick cryo-sections are transferred to a silicon ...

Comparison of three methods for mitochondria isolation from the human liver cell line (HepG2).

AIM The aim of this study was to evaluate and compare three available methods for mitochondrial isolation from a human cell line to predict the best method for each probable application. BACKGROUND Organelle isolation is gaining importance in experimental laboratory settings. Mitochondrial dysfunction may affect tumorgenesis process. There are some evidences that transplantation of healthy, i...

Use of dark ground microscopy in haematology.

The novel combination of vital staining, quick drying of thin films, and dark ground microscopy furnishes new information about organelles, granules, and precipitates in blood cells. The preparations are permanent and give optical signals of high contrast which depend on refraction or dichroism of the object. Mitochondria are readily shown and can be counted and sized. Pilot observations sugges...

O-21: Differential Expression and Epigenetic Pattern of HOX Family Genes in Cumulus Cells of Mature MII Oocytes from Patients with Polycystic Ovary Syndrome

Background Ovarian tissue cryopreservation represents a promising strategy to preserve the ovarian function in cancer patients. It is usually performed by slow freezing/rapid thawing (SF/RT). Recent studies emphasize an ultrarapid cryopreservation procedure, vitrification/warming (V/W), since it might prevent damages due to ice crystal formation. Comparative studies between the cryopreservation...
