چکیده

Previous autoradiographic and chemical studies gave incompatible results as regards the incorporation of cystine into and cystine content of the 2 cortical segments of the wool fibre. In this study the incorporation of ["S]cystine into the wool fibre cortex was therefore re-examined by electron-microscope autoradiography. Skin samples were taken from a Merino sheep 1 h and 5 h after intradermal injections of L-P'Sjcystine. At both times there was very little incorporation of S in the follicle bulbs. By 5 h incorporation occurred distally from the suprabulbar region throughout the zone of macrofibril (filament bundle) formation. More S was incorporated per unit area in the paracortex than in the orthocortex, and at the level of maximal uptake near the middle of this zone there was about a 2-fold difference per unit area. However, when the relative cross-sectional areas of the cortical segments were also considered, the actual amount of "S incorporated at this level was slightly greater in the orthocortex than in the paracortex. These differences in the incorporation of [S]cystine by the cortical segments agreed with previous results from chemical studies on cortical fractions separated from wool.

Incorporation of [Me-3H]methionine into wool follicle proteins: a biochemical and ultrastructural study.

The formation of the 'low-sulphur' group of proteins which constitutes more than half the cortical cell content of wool was investigated. Of the amino acids present in the lowand highsulphur proteins isolated from wool, methionine is the only one which is exclusive to the low-S group. The incorporation of [Me-'H]methionine, administered intradermally to a sheep, was examined using both biochemi...

An ultrastructural study of the membranes of keratinizing wool follicle cells.

Measurements of the widths of apposed plasma membranes and of the spaces between them were made at different stages of differentiation of keratinizing and hardening cells of the wool follicle. In contrast to findings in the hair follicle, no changes were detected for apposed cortex, cortex/fibre cuticle and fibre cuticle/fibre cuticle cells until keratinization had taken place. The trilaminar a...

The effect of increasing methionine supply on the methionine conversion to cyst(e)ine in sheep.

The conversion of methionine to cyst(e)ine was determined in sheep infused with different amounts of methionine (0-5 g/d) into the duodenum by assaying the incorporation of 35S from intravenously-infused L-[35S]methionine into cyst(e)ine in wool, plasma albumin and the free plasma pool. The percentage of cystine-S in the plasma originating from methionine increased linearly from 4.5 to 18 with ...

The Growth and Composition of Wool following Defleecing of Sheep with N-[5-(4-aminophenoxy)pentyl]phthalimide

In four sheep dosed with N-[5-(4-aminophenoxy)pentyl]phthalimide (400 mg/kg live weight) fibre growth stopped within l-ls days and the sheep were readily defleeced after 9 days. Most follicles stopped growing fibres for 12-16 days. When wool growth resumed, autoradiographic measurements indicated that it was slightly enhanced in the early regrowth due to an increase in fibre diameter. This incr...
