چکیده

Aim: To identify the Clostridium perfringens toxin genotypes prevailing in enterotoxemia suspected sheep flocks in Andhra Pradesh by using multiplex PCR. Materials and Methods: Intestinal scrapings were collected from lambs below three months of age from flocks with similar management from different Districts of Andhra Pradesh, in India. A total of 208 samples were collected with 140 from enterotoxemia suspected flocks and 68 from healthy flocks. Following processing and culture of the samples, colonies were identified by morphological and biochemical tests. All the clostridial isolates were analyzed by multiplex PCR. Results: C. pefringens were isolated from 97 out of 140 enterotoxemia suspected flocks (69.29%) and 27 out of 68 healthy flocks (39.71 %). Genotyping of the 97 iolates by multiplex PCR from enterotoxemia suspected flocks indicated C. perfringens type A, C and D 67.01% (65 out of 97); 11.34% (11 out of 97) and 21.65% (21 out of 97) respectively. Isolates from healthy flocks indicated the presence of type A and D 92.59% (25 out of 27) and 7.40% (2 out of 27) respectively. Number of isolates from enterotoxaemia suspected flocks were significantly high (P<0.001) than healthy flocks. Type A is found to be predominant in both enterotoxemia suspected and healthy flocks (67.01% and 92.59%). Conclusions: Prevalence of C. perfringens type C was reported for the first time in India. Clostridium perfringens type D and type C were found to be the major causative types for enterotoxemia.

Prevalence and Toxinotyping of the Toxigenic Clostridium perfringens in Sheep with suspected Enterotoxemia

The study was designed to determine the types of Clostridium perfringens and their toxins in sheep with suspected enterotoxemia in AlAhsa province, KSA. Out of 240 intestinal content samples collected from dead sheep with suspected enterotoxemia, 34 Clostridial strains were isolated. Diagnosis was based on classical diagnostic tests, enzyme-linked immunosorbent assay (ELISA) and multiplex polym...

Naturally acquired antibodies against Clostridium perfringens epsilon toxin in goats.

Clostridium perfringens type D-producing epsilon toxin is a common cause of death in sheep and goats worldwide. Although anti-epsilon toxin serum antibodies have been detected in healthy non-vaccinated sheep, the information regarding naturally acquired antibodies in ruminants is scanty. The objective of the present report was to characterize the development of naturally acquired antibodies aga...

Molecular typing of toxigenic Clostridum perfringens isolated from sheep in Iran

In this research a molecular method based on polymerase chain reaction for typing of Clostridium perfringens was developed and toxin genotypes of 64 isolates from sheep and goats in Iran were determined. The PCR assays were developed for detection of alpha (cpa), beta (cpb) and epsilon (etx) toxin genes, allowing classification of the isolates into genotypes A B, C and D. The field isolates ...

Cloning of Clostridium perfringens iota toxin gene in Escherichia coli

Iota toxin is produced by Clostridium perfringens type E. This toxin causes antibiotic-associated enterotoxemia in lambs and calves. Iota toxin is a binary toxin that has two components including Ia (the enzyme component) and Ib (the binding component). Ib binds to the surface receptor of target cells and translocate Ia into the cytosol of cells. The aim of this study was to clone toxigenic epi...

Toxinotyping of Clostridium perfringens isolates by ELISA and PCR from lambs suspected of enterotoxemia

Clostridium perfringens is a gram-positive, anaerobic bacterium that causes a wide range of diseases in humans and animals. Th e purposes of this study were to determine, using the enzyme-linked immunosorbent assay (ELISA), the clostridial toxins in intestinal samples of lambs with suspected enterotoxemia; to conduct molecular typing of C. perfringens isolates; and to investigate the presence o...
