Simple assay for monoacylglycerol hydrolase activity of rat adipose tissue.

چکیده
A simple, sensitive, and specific assay for monoacylglycerol hydrolase activity of rat adipose tissue is described. Monoacyl[(3)H]glycerols with different chain lengths (8-18 carbon atoms) and different degrees of unsaturation in mixed micellar solution with different detergents can be used as substrates. The [(3)H]glycerol that is produced is isolated in a one-step liquid-liquid partition procedure. For routine purposes monooleoyl[(3)H]glycerol was found to be the most suitable substrate. A simple method for the chemical synthesis and purification of this substrate in high yield is given. The assay allows rapid serial sampling of enzymatic activity with a high reproducibility.

Monoacylmonoalkylglycerol as a substrate for diacylglycerol hydrolase activity in adipose tissue.
The synthesis and use of 1(3)-[3H]oleoyl-2-0-oleylglycerol as a substrate for the assay of diacylglycerol hydrolase activity in adipose tissue is described. Neither the compound nor its reaction product are hydrolyzed by purified adipose tissue monoacylglycerol lipase.
متن کاملMesenchymal Stem Cells Derived from Rat Epicardial Versus Epididymal Adipose Tissue
Objective(s) Some investigation has indicated that adipose-derived stem cells possess different surface epitopes and differentiation potential according to the localization of fat pad from which the cells were derived. In the present study proliferation capacity and aging of such cells were explored. Materials and Methods Adherent cells were isolated from the collagenase digests of adipose tiss...
متن کاملOsteogenic Differentiation of Rat Mesenchymal Stem Cells from Adipose Tissue in Comparison with Bone Marrow Mesenchymal Stem Cells: Melatonin As a Differentiation Factor
Background: Adipose-derived stem cells (ADSC) could be an appealing alternative to bone marrow stem cells (BMSC) for engineering cell-based osteoinductive grafts. Meanwhile, prior studies have demonstrated that melatonin can stimulate osteogenic differentiation. Therefore, we assayed and compared the melatonin effect on osteogenic differentiation of BMSC with that of ADSC. Methods: Mesenchymal...
متن کاملDifferentiation of Adipose-derived Stem Cells into Schwann Cell Phenotype in Comparison with Bone Marrow Stem Cells
Objective(s) Bone marrow is the traditional source of human multipotent mesenchymal stem cells (MSCs), but adipose tissue appears to be an alternative and more readily available source. In this study, rat adipose-derived stem cells (ADSCs) were induced to differentiate into Schwann-like cells and compared with rat bone marrow stem cells (BMSCs) for their Schwann-like cells differentiation pote...
متن کاملCharacterization of the Mouse and Human Monoacylglycerol O-Acyltransferase 1 (Mogat1) Promoter in Human Kidney Proximal Tubule and Rat Liver Cells
Monoacylglycerol acyltransferase 1 (Mogat1) catalyzes the conversion of monoacylglycerols (MAG) to diacylglycerols (DAG), the precursor of several physiologically important lipids such as phosphatidylcholine, phosphatidylethanolamine and triacylglycerol (TAG). Expression of Mogat1 is tissue restricted and it is highly expressed in the kidney, stomach and adipose tissue but minimally in the norm...
