چکیده

Morita, Richard Y. (Oregon State University, Corvallis), and Paul F. Mathemeier. Temperature-hydrostatic pressure studies on partially purified inorganic pyrophosphatase activity. J. Bacteriol. 88:1667-1671. 1964.-Partially purified inorganic pyrophosphatase from Bacillus stearothermophilus demonstrated increased activity at 90 C with hydrostatic pressures of 100 to 1,100 atm. At 90 C, optimal activity was at 700 atm. No enzyme activity could be demonstrated at 100 C at 1 atm; however, at 105 C moderate hydrostatic pressures favored the enzyme reaction, resulting in increased activity. The increased enzyme activity is explained on the basis of pressure counteracting the molecular volume increase which results from elevated temperature. Thermal denaturation studies at 90 C show inorganic pyrophosphatase to be more stable in the presence of cofactor than substrate, and indicate an enyzme-cofactor intermediate in the hydrolysis.

Purification of yeast inorganic pyrophosphatase.

An enzyme which hydrolyzes inorganic pyrophosphate (PP) to orthophosphate (ortho P) was demonstrated by Lohmann (1) in muscle. Bauer (2) found that autolysates of slowly dried bottom yeast were a rich source of inorganic pyrophosphatase and were free of adenosinetriphosphatase (ATPase) activity. An important contribution was made by Bailey and Webb (3) who accomplished an extensive purification...

Effects of hydrostatic pressure and temperature on the AlGaN/GaN High electron mobility transistors

In this paper, drain-source current, transconductance and cutoff frequency in AlGaN/GaN high electron mobility transistors have been investigated. In order to obtain parameters of exact AlGaN/GaN high electron mobility transistors such as electron density, the wave function, band gap, polarization charge, effective mass and dielectric constant, the hydrostatic pressure and temperature effects a...

Purification and some properties of inorganic pyrophosphatase from human erythrocytes.

Inorganic pyrophosphatase was purified BOO-fold from human erythrocytes by a procedure involving complete hemolysis of the cells, removal of hemoglobin, ammonium sulfate fractionation, diethylaminoethyl cellulose column chromatography, and gel filtration. Magnesium chloride and 2-mercaptoethanol stabilized the activity. The procedure yielded an enzyme with an optimal pH of 7.7, and an apparent ...

Subunit interaction of vacuolar H+-pyrophosphatase as determined by high hydrostatic pressure.

Vacuolar H+-pyrophosphatase (H+-PPase) from etiolated hypocotyls of mung bean (Vigna radiata L.) is a homodimer with a molecular mass of 145 kDa. The vacuolar H+-PPase was subjected to high hydrostatic pressure to investigate its structure and function. The inhibition of H+-PPase activity by high hydrostatic pressure has a pressure-, time- and protein-concentration-dependent manner. The Vmax va...

Inorganic pyrophosphatase is a component of the Drosophila nucleosome remodeling factor complex.

The Drosophila nucleosome remodeling factor (NURF) is a protein complex consisting of four polypeptides that facilitates the perturbation of chromatin structure in vitro in an ATP-dependent manner. The 140-kD NURF subunit, imitation switch (ISWI), is related to the SWI2/SNF2 ATPase. Another subunit, NURF-55, is a 55-kD WD repeat protein homologous to the human retinoblastoma-associated protein ...
