Nutritional requirements for Hydrogenomonas eutropha.

چکیده
Repaske, Roy (National Institute of Allergy and Infectious Diseases, Bethesda, Md.). Nutritional requirements for Hydrogenomonas eutropha. J. Bacteriol. 83: 418-422. 1962.-A simple apparatus for the autotrophic cultivation of Hydrogenomonas eutropha in 100-ml shake cultures is described. Nitrogen, in the form of ammonium, nitrate, or urea, was used for growth; nitrite could not be utilized. Optimal growth occurred at pH 6.4 to 6.8 at 30 C. H. eutropha grew best in an atmosphere containing 15 to 25% oxygen and 10% carbon dioxide. Below these concentrations each of the gases was limiting. Growth was shown to be dependent on iron, and the rate of growth was a function of iron concentration and its state of oxidation.

Yields of Hydrogenomonas eutropha from Growth
Molar growth yields were determined from chemostat cultures of Hydrogenomonas eutropha on succinate and on fumarate. The yields from culture on succinate were about 12 g higher than on fumarate. Assuming this difference to be equivalent to 1 molecule of adenosine triphosphate, it is concluded that the oxidation by oxygen of the Hydrogenomonas cytochrome b yields 1 molecule of adenosine triphosp...
متن کاملFactors affecting the synthesis and degradation of ribulose-1,5-diphosphate carboxylase in Hydrogenomonas facilis and Hydrogenomonas eutropha.
Hydrogenomonas facilis and H. eutropha cultured in fructose medium retained high levels of ribulose-1,5-diphosphate carboxylase only when the following conditions were fulfilled: low aeration, FeCl(3) addition to fructose medium, and cell harvest at or prior to mid-exponential phase of growth. Repression of carboxylase synthesis was demonstrated under conditions of high oxygen tension during gr...
متن کاملUtilization of amino acids as a source of nitrogen by Hydrogenomonas eutropha.
Relatively little has been published on the capacity of Hydrogenomonas to utilize amino acids under conditions supporting either autotrophic or heterotrophic growth. DeCicco and Umbreit (4) found that wild-type H. facilis utilizes either phenylalanine or tyrosine for growth with glucose as the main carbon and energy source. Atkinson (2) noted that H. facilis growing autotrophically in the prese...
متن کاملThe Electron Transport System of Hydrogenomonas eutropha
Wosilait and Nason (1) demonstrated that quinones could serve as electron acceptors for the enzymatic oxidation of reduced pyridine nucleotides, and their survey of plant, animal, and microorganism extracts (2) revealed the general occurrence of reduced pyridine nucleotide-quinone reductase activity. This finding and Dam’s (3) earlier report, which indicated that vitamin K was a common constitu...
متن کاملpH-conditional, ammonia assimilation-deficient mutants of Hydrogenomonas eutropha: evidence for the nature of the mutation.
Two amination-deficient mutants of Hydrogenomonas eutropha, characterized by pH-dependent linear growth on non-amino acid substrates, were investigated to determine the exact nature of the mutation. Glutamate dehydrogenase, the only aminating enzyme found in wild-type cells, was present at similar levels in mutant cells. Phenylalanine and aspartate, which allowed normal growth of the mutants, c...
