Kinetics of Trypsin Digestion

چکیده

The anomalies of enzyme action have frequently been accounted for by the assumption of an intermediate compound between the enzyme and the substrate. I t was shown, however, by Nelson and Hitchcock t that in the case of invertase the experiments could not be accurately calculated on this basis. The writer found with trypsin' that although the effect of varying the substrate concentration could be accounted for, the effect of varying both the substrate and inhibiting substances could not, provided the law of mass action was assumed to hold. The question is one of such importance from the view of all enzyme reactions, however, that it seems important to obtain direct experimental evidence in regard to the existence of such a compound. The statement is frequently made that the substrate protects the enzyme from heat inactivation and this has been considered as evidence for the existence of a compound between the substrate and enzyme. I t has been found, 3 however, that this protective action does not become manifest at once but only after the enzyme and substrate have been in contact, when it becomes more and more pronounced. I t can further be shown that the products of digestion afford very marked protection and that this effect is apparent at once. I t appears probable, therefore, that the protective action of the substrate solution is not due to the substrate itself but to the products formed by the action of the enzyme and that this is the reason for the delay in the protective action.

Quantitative proteomics reveals the kinetics of trypsin-catalyzed protein digestion.

Trypsin is the popular protease to digest proteins into peptides in shotgun proteomics, but few studies have attempted to systematically investigate the kinetics of trypsin-catalyzed protein digestion in proteome samples. In this study, we applied quantitative proteomics via triplex stable isotope dimethyl labeling to investigate the kinetics of trypsin-catalyzed cleavage. It was found that try...

The kinetics of trypsin digestion.

The present study of a typical proteolytic enzyme extends the quantitative investigations previously reported from this laboratory on the effects of pressure on protein systems in viva (1) and in vitro (2). This paper presents new data on the self-digestion of trypsin and the digestion of casein by trypsin; a forthcoming paper will discuss the effects of temperature and hydrostatic pressure on ...

Trypsin and chymotrypsin are necessary for in vitro enzymatic digestion of rice starch

An appropriate in vitro starch digestion model is necessary to comparatively evaluate the digestibility of starch samples and to help predict the in vivo glycemic responses of starchy foods. In the present study, the effect of trypsin and chymotrypsin on in vitro digestion of rice starch was studies for the first time. Inclusion of the trypsin and chymotrypsin in the digestion protocols, to mor...

A Recyclable Poly(ionic liquid)s Enzyme Reactor for Highly Efficient Protein Digestion

One of the most significant tasks for proteomic research and industrial applications, is the preparation of recyclable enzyme reactor. Herein, a novel recyclable enzyme reactor has been developed based on monodispersed spherical poly(quaternary ammonium ionic liquid)s particles immobilized trypsin. A new quaternary ammonium ionic liquids functional monomer was first synthesized. The ionic l...

Use of self assembled magnetic beads for on-chip protein digestion.

The use of grafted trypsin magnetic beads in a microchip for performing protein digestion is described. The PDMS device uses strong magnets to create a magnetic field parallel to the flow with a strong gradient pointing through the center of the chip channel. This allows for the formation of a low-hydrodynamic resistance plug of magnetic trypsin beads that serves as a matrix for protein digesti...

Digestion kinetics of carbohydrate fractions of citrus by-products

The present experiment was carried out to determine the digestion kinetics of carbohydrate fractions of citrus by-products. Grapefruit pulp (GP), lemon pulp (LE), lime pulp (LI) and orange pulp (OP) were the test feed. Digestion kinetic of whole citrus by-products and neutral detergent fiber (NDF) fraction and acid detergent fiber (ADF) fractions of citrus by-products were measured using the <e...
