Structural studies of capsular polysaccharide of type XII Diplococcus pneumoniae.

چکیده
The capsular polysaccharide of Diplococctw pneumoniae Type XII contains residues of D-glucose and D-galactose in a molar ratio of 2:l. The methylated polysaccharide yielded upon hydrolysis 2,3,4,6-tetraand 3,4,6-tri-O-methyl-D-glucose and 2,3,4,6-tetra-0-methyl-D-galactose as the only neutral methyl sugars. Periodate oxidation of the polysaccharide resulted in destruction of all neutral sugars and immunochemical activity against rabbit antisera. Periodate oxidation of the methyl 0-methylglycosides obtained after hydrolysis of the methyiated polysaccharide indicated that at least 30% of the L-fucosamine residues are substituted at C-4 in the polysaccharide. It is concluded that the polysaccharide consists of a hexosamine backbone that is substituted by D-galactosyl and kojibiosyl side-chains. The proposed terminal D-galactosyl residues apparently are sterically hindered from interacting with several D-galactose-binding proteins.

Filamentous capsulated streptococci from the human respiratory tract. I. Antigenic attributes of provisional capsular type 83 and its relationship to streptococci of so-called group M.
Two immunologically reactive polysaccharides have been isolated from the cell walls and from culture filtrates of a filamentous alpha-hemolytic streptococcus provisionally designated capsular type 83. Both polysaccharides were purified by diethylaminoethyl-cellulose chromatography. Analysis indicates that the capsular polysaccharide consists of galactose and phosphorus, whereas the cell wall po...

Abstract Background and objectives: important virulence factor for many invasive bacterial pathogens of humans. Escherichia coli offer a model system to study the mechanisms by which capsular polysaccharides are synthesized and exported onto the cell surface of bacteria. Biosynthesis of the E consists of the repeat structure -4) GlcA- (1, 4)-GlcNAc- (1-, requires the KfiA,...
متن کاملIdentification of Capsular Serotypes K1 and K2 in Clinical Isolates of Klebsiella Pneumoniae in North of Iran
ABSTRACT Background and Objectives: Klebsiella pneumoniae is one of the most important nosocomial pathogens. Its capsular polysaccharide is considered as the first and most important virulence factor of this bacterium. This study aimed to investigate the presence of capsular serotypes K1 and K2 in K. pneumoniae isolates to examine the virulence ...
متن کاملThe pathogenicity of Klebsiella pneumoniae for mice: the relationship to the quantity and rate of production of type-specific capsular polysaccharide.
It is well established that among certain groups of microorganisms the presence of a capsule is essential for virulence; these studies are largely limited to all-or-none effects, i. e. the presence or absence of capsule. Among a few species, e. g. Diplococcus pneumoniae, some quantitative studies have been made which indicate that there is a relationship between size of capsule and degree of pa...
متن کاملCharacterization of the cassette containing genes for type 3 capsular polysaccharide biosynthesis in Streptococcus pneumoniae
The capsular polysaccharide is the major virulence factor of Streptococcus pneumoniae. Previously, we identified and cloned a region from the S. pneumoniae chromosome specific for the production of type 3 capsular polysaccharide. Now, by sequencing the region and characterizing mutations genetically and in an in vitro capsule synthesis assay, we have assigned putative functions to the products ...
