Small Molecule Inhibitors of Ab-Aggregation and Neurotoxicity
نویسندگان
چکیده
Alzheimer disease (AD) is characterized pathologically by extracellular amyloid deposits composed of Ab peptide, neurofibrillary tangles (NFTs) made up of hyperphosphorylated tau, and a deficit of cholinergic neurons in the basal forebrain. Presently, only symptomatic therapies are available for the treatment of AD and these therapies have a limited time frame of utility. Amyloid disorders represent the effects of chronic Ab production and are not a secondary pathological effect caused by a distant trigger; therefore targeting Ab is a viable pursuit. In this review, we will discuss the various small molecule antiaggregation inhibitors that have been reported in the literature, with emphasis on compounds that are presently being investigated in clinical trials. متن کاملMyricetin: a naturally occurring regulator of metal-induced amyloid-β aggregation and neurotoxicity.
One of the most severe and incurable forms of neurodegeneration, Alzheimer’s disease (AD), is characterized in the brain by the accumulation of aggregated amyloid-b (Ab) peptides. In the diseased brain, elevated concentrations of metals, such as Fe, Cu, and Zn, are found in Ab plaques. It has been proposed that metal ions, such as Cu and Zn, can bind to Ab ; this causes enhanced peptide aggrega...
متن کاملA Sensitive A Oligomerization Assay for Identification of Small Molecule Inhibitors
Amyloid deposits found in Alzheimer’s disease result from aggregation of peptide which leads to loss of synaptic function, chronic microglial activation and cognitive impairment. Because of this, identification of small molecule inhibitors of aggregation as potential therapeutics is a topic of current interest. The majority of inhibitor screening approaches rely on in vitro assays that lack the...
متن کاملHigh-Content Chemical and RNAi Screens for Suppressors of Neurotoxicity in a Huntington's Disease Model
To identify Huntington's Disease therapeutics, we conducted high-content small molecule and RNAi suppressor screens using a Drosophila primary neural culture Huntingtin model. Drosophila primary neurons offer a sensitive readout for neurotoxicty, as their neurites develop dysmorphic features in the presence of mutant polyglutamine-expanded Huntingtin compared to nonpathogenic Huntingtin. By tra...
متن کاملMolecular Dynamics and Molecular Docking Studies on the Interaction between Four Tetrahydroxy Derivatives of Polyphenyls and Beta Amyloid
Interactions of 3,3',4,4'-tetrahydroxybiphenyl (BPT) and three isomeric 3,3",4,4"-tetrahydroxyterphenyls (OTT, MTT, PTT) with Alzheimer’s amyloid-β peptide (Aβ) were studied by molecular dynamics simulation and molecular docking. Structural parameters such as Root-mean-square derivations (RMSD), radial distribution function (RDF), helix percentage and other physical parameters were obtained. Th...
