چکیده

Bacterial cold water disease (BCWD) in ayu (Plecoglossus altivelis altivelis) has a serious impact on aquaculture and fisheries. There is known to be a significant difference among ayu strains with regard to mortality caused by BCWD. In this study, the immune response of different ayu strains against Flavobacterium psychrophilum infection was observed. One strain was resistant to infection by F. psychrophilum, and the other was susceptible to infection by the same bacteria. The number of bacteria in the body was observed in each ayu strain, and the change in bacterial counts was similar. However, there was a significant difference in bacterial count in the spleen between the two strains on days 6, 9, 12 and 15 after exposure. To observe the immune response against F. psychrophilum, agglutination assay using serum was performed. An agglutination reaction in the resistant ayu strain was observed in 4 out of 6 ayu on day 6 after exposure, while no reactions in the susceptible ayu strain were observed in any sampled fish until day 12. However, some reactions in the susceptible ayu strain were observed in surviving ayu. These results indicate that there is a correlation between the presence of bacterial multiplication and agglutination reaction against F. psychrophilum.

Plasmid profiling of Flavobacterium psychrophilum isolates from ayu (Plecoglossus altivelis altivelis) and other fish species in Japan

In order to evaluate the genetic variability of the causative agent of cold water disease (CWD), plasmid profiling was used to characterize Flavobacterium (F.) psychrophilum isolates (n = 169). Size analysis of plasmids in F. psychrophilum isolates (n = 128) from several fish species demonstrated that six kinds of plasmids were harbored, and ayu isolates had different profiles compared to other...

Pathogenesis of experimentally induced bacterial cold water disease in ayu Plecoglossus altivelis.

Ayu Plecoglossus altivelis were experimentally infected with Flavobacterium psychrophilum, which is the causative agent of bacterial cold water disease (CWD). The fish infected by immersion usually died within an hour after they became moribund. The blood volume and haematocrit values of moribund fish were low, while those values of many infected fish that were not moribund were in the range of...

Draft Genome Sequence of Flavobacterium psychrophilum Strain SSADA-1411, Isolated from an Ayu (Plecoglossus altivelis altivelis) Migrating Downriver To Spawn in the Shimanto River, Kochi, Japan

Here, we report the draft genome sequence and annotation of Flavobacterium psychrophilum strain SSADA-1411. This strain was isolated from the skin ulcer of an ayu (Plecoglossus altivelis altivelis) migrating downriver to spawn in the lower Shimanto River, in western Kochi Prefecture on Shikoku Island in Japan.

Nation-wide and local diversity of the fish pathogen Flavobacterium psychrophilum in Japan revealed by MLST analysis

Flavobacterium psychrophilum is the causative agent of bacterial cold-water disease (BCWD) and rainbow trout fry syndrome in salmonid fish. In Europe and in North and South America, F. psychrophilum usually affects salmonid hosts such as rainbow trout (Oncorhynchus mykiss) and coho salmon (Oncorhynchus kisutch). However, in Japan, ayu (Plecoglossus altivelis altivelis) are more frequently affec...

Genotyping of Flavobacterium psychrophilum isolates from wild and farmed salmonids in Norway and Chile

Flavobacterium psychrophilum, a member of the family Flavobacteriaceae, has from the first descriptions in North America, been distributed to many countries worldwide causing serious losses to the aquaculture industry. The Norwegian and Chilean aquacultures are also affected by outbreaks due to F. psychrophilum. The bacterium has been one of the most common causes of mortality in fresh water aq...
