چکیده

Pregnane and Xenobiotic Receptor (PXR), a vital xenosensor, acts as master regulator of phase-I (cytochrome P450) and phase-II enzymes (glutathione S-transferases, sulfotransferases, and uridine 5’-diphosphate glucuronosyltransferases) as well as several drug transporters (multi-drug resistance protein, and multidrug resistance-associated protein). PXR can bind to a variety of chemically distinct endobiotics (steroids, bile acids and their derivatives, vitamins, etc.) and xenobiotics (prescription drugs, herbal medicines, endocrine disruptors, etc.). Activation of PXR by various compounds leads to trans-activation of PXRtarget genes involved in detoxification system (phase-I and phase-II enzymes, and efflux proteins). Herbal medicines are readily used without prescription under the belief that anything natural is safe. These medicines contain active chemical constituents which execute distinctly different or similar pharmacological response(s). But, like prescription drugs, herbal drugs also have both therapeutic and, sometimes, adverse effects. Some of the herbal drugs induce drug metabolizing enzymes (especially CYP3A4) and drug efflux proteins via activation of PXR. Phase-I enzyme CYP3A4 is involved in the metabolism of 50-60% of clinical drugs as well as the chemical ingredients in herbal medicines. In addition to this, 25-30% of these compounds are metabolized by the CYP2B isoenzymes. The combined metabolic effects of phase-I and phase-II enzymes and drug transporters, following induction by therapeutic molecules, constitute the molecular basis for many drug-herbal interactions. For example, if one drug activates PXR, it can encourage the elimination of a co-administered drug that is also metabolized and eliminated by PXR-target gene products, thereby affecting the therapeutic efficacy of the drug in the context of combination therapy. The present review highlights some of the recent clinical correlates in drug-herbal interactions mediated primarily via PXR and cytochrome P450.

Induction of cytochrome P450 3A by paclitaxel in mice: pivotal role of the nuclear xenobiotic receptor, pregnane X receptor.

Paclitaxel, a taxane anti-microtubule agent, is known to induce CYP3A in rat and human hepatocytes. Recent studies suggest that a member of the nuclear receptor family, pregnane X Receptor (PXR), is a key regulator of the expression of CYP3A in different species. We investigated the role of PXR activation, in vitro and in vivo, in mediating Cyp3a induction by paclitaxel. Pregnenolone 16 alpha-c...

Resveratrol suppresses the inducible expression of CYP3A4 through the pregnane X receptor.

The pregnane X receptor (PXR, NR1I2), a member of the nuclear receptor superfamily, is activated by a number of clinically prescribed drugs and herbal extracts. The inducible expression of several important cytochrome P450 (CYP450) enzymes has been shown to be regulated by the activation of PXR in the liver. In the current study, reporter gene-transfected cells were used to identify potential a...

Utility of DPX2 cells for predicting CYP3A induction-mediated drug-drug interactions and associated structure-activity relationships.

The increase in cytochrome P450 (P450) enzyme activity noted upon exposure to therapeutics can elicit marked drug-drug interactions (DDIs) that may ultimately result in poor clinical outcome or adverse drug effects. As such, in vitro model systems that can rapidly and accurately determine whether potential therapeutics activate the human pregnane X receptor (PXR) and thus induce CYP3A P450 leve...

Glucocorticoid-mediated induction of CYP3A4 is decreased by disruption of a protein: DNA interaction distinct from the pregnane X receptor response element.

CYP3A4 is the most abundant cytochrome P450 (P450) in human liver, comprising approximately 30% of the total liver P450 content. This enzyme has an important role in steroid catabolism and metabolism of foreign compounds, with the majority of pharmaceutical compounds being substrates for CYP3A4. The molecular mechanisms that underlie transcriptional activation of CYP3A4 are complex with many st...
