Binding of copper(I) by the Wilson disease protein and its copper chaperone.

The Wilson disease protein (WND) is a transport ATPase involved in copper delivery to the secretory pathway. Mutations in WND and its homolog, the Menkes protein, lead to genetic disorders of copper metabolism. The WND and Menkes proteins are distinguished from other P-type ATPases by the presence of six soluble N-terminal metal-binding domains containing a conserved CXXC metal-binding motif. T...

Survey of heavy metal (Copper, Iron, Lead, Cadmium, Zinc and Nickel) concentrations and their effects on the water quality of Anzali wetland

Anzali International wetland is one of the most valuable water resources as registered in Ramsar convention with an extent of about 150 km2 in the south Caspian Sea. This wetland, not only has ecological and biological importance for migratory birds and rare fish species, but also is used for farms to discharge into the Caspian Sea through this wetland. Therefore its pollution control is very ...

A new zinc-protein coordination site in intracellular metal trafficking: solution structure of the Apo and Zn(II) forms of ZntA(46-118).

Zinc, a metal ion that functions in a wide variety of catalytic and structural sites in metalloproteins, is shown here to adopt a novel coordination environment in the Escherichia coli transport protein ZntA. The ZntA protein is a P-type ATPase that pumps zinc out of the cytoplasm and into the periplasm. It is physiologically selective for Zn(II) and functions with metalloregulatory proteins in...

Thermodynamics of copper and zinc distribution in the cyanobacterium Synechocystis PCC 6803.

Copper is supplied to plastocyanin for photosynthesis and cytochrome c oxidase for respiration in the thylakoids of Synechocystis PCC 6803 by the membrane-bound P-type ATPases CtaA and PacS, and the metallochaperone Atx1. We have determined the Cu(I) affinities of all of the soluble proteins and domains in this pathway. The Cu(I) affinities of the trafficking proteins range from 5 × 10(16) to 5...

A P-type ATPase importer that discriminates between essential and toxic transition metals.

Transition metals, although being essential cofactors in many physiological processes, are toxic at elevated concentrations. Among the membrane-embedded transport proteins that maintain appropriate intracellular levels of transition metals are ATP-driven pumps belonging to the P-type ATPase superfamily. These metal transporters may be differentiated according to their substrate specificities, w...
