چکیده

1. Simultaneous and continuous measurements of changes in CO2 and 02 concentrations in glucose-metabolizing yeast suspensions by mass spectrometry enabled a study of the Pasteur effect (aerobic inhibition of glycolysis) in Saccharomyces uvarum and Schizosaccharomyces pombe. 2. A different control mechanism operates in Candida utilis to give a damped oscillation after the anaerobic-aerobic transition. 3. The apparent Km values for respiration of the three yeasts were in the range 1.3-1.8 pM-02. 4. The apparent Km values for 02 of the Pasteur effect were 5 and 13 pM for catabolite-repressed and derepressed S. uvarum respectively and 7 UM for Sch. pombe. 5. These results are discussed with respect to currently accepted mechanisms for the control of glycolysis.

Mathematical models for explaining the Warburg effect: a review focussed on ATP and biomass production.

For producing ATP, tumour cells rely on glycolysis leading to lactate to about the same extent as on respiration. Thus, the ATP synthesis flux from glycolysis is considerably higher than in the corresponding healthy cells. This is known as the Warburg effect (named after German biochemist Otto H. Warburg) and also applies to striated muscle cells, activated lymphocytes, microglia, endothelial c...

Phosphorylation of mononucleotides and formation of cytidine 5'-diphosphate-choline and sugar nucleotides by respiration-deficient mutants of yeasts.

Respiration-deficient mutants (Rho-, petite) of Saccharomyces carlsbergensis were obtained by treatment with trypaflavin (euflavine). Dried cells of these mutants phosphorylated mononucleotides to their triphosphates and further formed not only cytidine 5'-diphosphate-choline, but also sugar nucleotides, such as uridine 5'-diphosphate-glucose, guanosine 5'-diphosphate-mannose, etc. The activiti...

Carbohydrate and energy-yielding metabolism in non-conventional yeasts.

Sugars are excellent carbon sources for all yeasts. Since a vast amount of information is available on the components of the pathways of sugar utilization in Saccharomyces cerevisiae it has been tacitly assumed that other yeasts use sugars in the same way. However, although the pathways of sugar utilization follow the same theme in all yeasts, important biochemical and genetic variations on it ...

Involvement of Cytochrome P-450 in n-Butyl Nitrite-Induced Hepatocyte Cytotoxicity

Addition of n-butyl nitrite to isolated rat hepatocytes caused an immediate glutathione depletion followed by an inhibition of mitochondrial respiration, inhi- bition of glycolysis and ATP depletion. At cytotoxic butyl nitrite concentrations, lipid  peroxidation  occurred  before  the  plasma  membrane  was  disrupted. Cytochrome P-450 inhibitors inhibited peroxynitrite formation and prev...

Gas bubble formation in the cytoplasm of a fermenting yeast

Current paradigms assume that gas bubbles cannot be formed within yeasts although these workhorses of the baking and brewing industries vigorously produce and release CO(2) gas. We show that yeasts produce gas bubbles that fill a significant part of the cell. The missing link between intracellular CO(2) production by glycolysis and eventual CO(2) release from cells has therefore been resolve...
