چکیده
LTHOUGH fumarase was discovered by Einheck1 in 1919 and has subsequently been found to have a wide biological distribution, there are no reports to our knowledge of its activity in separated human beukocvtes. Fitmarase is one of the essential enzymes that catalyze the oxidation of carbohydrates, fatty acids and some amino acids via the citric acid cycle. Specifically, fumarase mediates the establishment of an equilibrium between fumaric acid and b-mabic acid.2 Because of the importance of this enzyme in cellular metabolism and the ease of “biopsying” the blood, it seemed pertinent to assay fitmarase activity in the leukocytes in leukemia as well as in other diseases. THE ISOLATION OF ENZYME TRANSKETOLASE FROM HUMAN ERYTHROCYTES: THE CHARACTERIZATION OF ITS QUARTERNARY STRUCTURE
Human erythrocyte transketolase (sedoheptulose-7-phosphate: D-glyceraldehyde-3-phosphate, glycolaldehyde transferase, E.C. 2.2.1.1.) has been isolated from erythrocytes with a specific activity of 59.84 U/mg. SDS-PAGE and SE-HPLC were used both as a measure of purity and as a preparative mean to obtain a higher degree of purity. Four protein bands corresponding to molecular weights of 32,0...
متن کاملBiochemical Characterization of Two Clinically-Relevant Human Fumarase Variants Defective for Oligomerization
Background Fumarase, a significant enzyme of energy metabolism, catalyzes the reversible hydration of fumarate to L-malate. Mutations in the FH gene, encoding human fumarase, are associated with fumarate hydratase deficiency (FHD) and hereditary leiomyomatosis and renal cell cancer (HLRCC). Fumarase assembles into a homotetramer, with four active sites. Interestingly, residues from three of the...

Background & Aim: The biological effects of potent oxidative agents in human body are under anti-oxidative control. Functional defects of organs may result from reactions of free radicals with the cell membrane. It is known that major targets of oxygen radicals are cell membrane lipids. Some reports indicate the role of peroxides in development of atherosclerosis. Human body tissues(eg.eryt...
متن کاملINHIBITION OF HUMAN ERYTHROCYTE GLUCOSE 6-PHOSPHATE DEHYDROGENASE ACTIVITY BY DEHYDROEPIANDROSTERONE AND RELATED STEROIDS.
The inhibitory effects of several steroids on G6PD activity using intact erythrocytes are reported. Incubation of whole blood with dehydroepiandrosterone (DHEA) resulted in 42% and 12% inhibition in the enzyme activity in the presence and absence of oxygen, respectively. Addition of epinephrine and/or aminophylline into the incubation medium caused further enzyme inhibition suggesting a po...
متن کاملIn vitro Antioxidant, Cytotoxic, Cholinesterase Inhibitory Activities and Anti-Genotoxic Effects of Hypericum retusum Aucher Flowers, Fruits and Seeds Methanol Extracts in Human Mononuclear Leukocytes
The present study investigates the antioxidant, anticancer, anticholinesterase, anti-genotoxic activities and phenolic contents of flower, fruit and seed methanol extracts of Hypericum retusum AUCHER. The amounts of protocatechuic acid, catechin, caffeic acid and syringic acid in methanol extracts were determined by HPLC. Total phenolic content of H. retusum seed extract was found more than fru...
