چکیده

Here we discuss a recently published study from our group on how a continuous CD4 T-cell dependent immune clearance of premalignant senescent cells, designated "senescence surveillance," restricts liver cancer development.

Registered report: Senescence surveillance of pre-malignant hepatocytes limits liver cancer development

The Kang et al. (2011), published in Nature in 2011. The experiments that will be replicated are those reported in Figures 3B, 3C, 3E, and 4A. In these experiments, Kang et al. (2011) demonstrate the phenomenon of oncogene-induced cellular senescence and immune-mediated clearance of senescent cells after intrahepatic injection of NRAS (Figures 2I, 3B, 3C, and 3E). Additionally, Kang et al. (201...

The usual SASPects of liver cancer

senescence is a stable form of cell cycle arrest that limits the propagation of damaged cells and can be triggered in response to diverse forms of cellular stress [1]. This anti-proliferative program is mediated by the p53 and Rb pathways and was initially considered a cell-autonomous mechanism that promotes tumor suppression and tissue homeostasis [1]. However, several groundbreaking studies p...

Clearance of senescent hepatocytes in a neoplastic-prone microenvironment delays the emergence of hepatocellular carcinoma

Increasing evidence indicates that carcinogenesis is dependent on the tissue context in which it occurs, implying that the latter can be a target for preventive or therapeutic strategies. We tested the possibility that re-normalizing a senescent, neoplastic-prone tissue microenvironment would exert a modulatory effect on the emergence of neoplastic disease. Rats were exposed to a protocol for t...

Regulation of -Fetoprotein by Nuclear Factor- B Protects Hepatocytes from Tumor Necrosis Factor- Cytotoxicity during Fetal Liver Development and Hepatic Oncogenesis

Nuclear factorB (NFB) plays a critical role during fetal liver development and hepatic oncogenesis. Here, we have assessed whether NFB activity is required for murine hepatocellular carcinoma cell survival. We show that adenoviral-mediated inhibition of inhibitor of NFB kinase(IKK-2) activity in hepatocellular carcinomas derived from transforming growth factor (TGF)/c-myc bitransgenic mice lead...

Hepatitis B virus down-regulates expressions of MHC class I molecules on hepatoplastoma cell line.

Chronic HBV infection is associated with a 100-fold high risk of developing hepatocellular carcinoma. Tumor recognition is of the most importance during the immune surveillance process that prevents cancer development in humans. In the present study, the expressions of MHC class I molecules on hepatoplastoma cell line HepG2.2.15 were investigated to indicate the possible effects of HBV on the i...
