Quantitative Assessment of Trimethyltin Induced Pathology
نویسندگان
چکیده
The pathology induced by trimethyltin in the hippocampus was investigated using a detailed morphometric model. Male Long-Evans rats were dosed with 3 mdkg trimethyltin chloride (TMT) for three consecutive days and sacrificed at subsequent interim periods. Total brain volume was not significantly affected; hippocampus volume decreased within 4 days after completion of dose. Pyramidal cells were more sensitive to the toxic effects of TMT than granule cells, in addition, dorsal pyramidal cells appeared to be more sensitive than ventral pyramidal cells. The dorsal CA4 region appeared to be the most susceptible to cell loss, although thc dor\sal CAI region also exhibited significant reductions. The most dramatic reduction in cell numbers occurred between 14 and 28 days post-dose. Cell loss within the CA3c and CA4 but not the CAI region was preceded by decreased nuclear volume. In addition cellular loss within the dorsal CAI region appeared to be most pronounced within a well defined rectangular patch of cells immediately adjacent to the CA3-CA1 junction. The data further suggest that TMT is a potent toxicant to the hippocampus. This toxicity is delayed, and selectively affects well defined groups of cells within the structure. Additionally, differences in nuclear pathology suggest that more than one mechanism may be involved in the destruction of these target cell populations.

Morphometric analysis of the developing rat brain.
In 2 studies, a method of linear morphometry was applied to regulatory developmental neurotoxicity studies in the rat. The first study involved the development of the brain during postnatal days (PNDs) 7-63, and the second involved the effects of 8 mg/kg i.p. trimethyltin chloride (TMT) to rats at PND 8, with morphometry performed at PNDs 12 and 24. The results of the TMT linear morphometry wer...

Background & aim: Given that Royal Jelly contains compounds that have beneficial effects on the central nervous system and neural functions, the aim of this study was to investigate the effect of eight weeks of Royal Jelly consumption on some of apoptotic characteristics in trimethyltin-induced Alzheimerchr('39')s rats Methods: The present study was performed experimentally with 32 male rats; T...
متن کاملChanges in muscarinic (M1 and M2 subtypes) and phencyclidine receptor density in the rat brain following trimethyltin intoxication.
The main purpose of this study was to determine whether one or both of the muscarinic receptor subtypes (M1 and M2) contributed to the total cholinergic receptor loss found in trimethyltin (TMT) treated rats and to assess the effect of TMT on phencyclidine (PCP) receptor density in several regions of the rat brain. The distribution and changes in muscarinic (M1 and M2) receptor and PCP receptor...
متن کاملPeripheral and central nervous system lesions caused by triethyl- and trimethyltin salts in rats.
Both trimethyltin and triethyltin salts are known to produce toxic lesions in the central nervous system. Triethyltin intoxication has been associated with central intramyelin edema, while trimethyltin has been shown to produce neuronal necrosis in selected limbic and sensory regions of the brain. Only scant attention has been paid to peripheral nerves of animals treated with alkyltins. In this...
متن کاملPentoxifylline Protects Against Hippocampal Damage and Memory Impairment Induced by Trimethyltin
Background: Trimethyltin (TMT) is a toxic agent that causes oxidative stress, a laboratory model for inducing hippocampal injuries. Pentoxifylline (PTX) inhibits phosphodiesterase, inflammation and oxidative stress. This study evaluated the neuroprotective effects of PTX on injuries induced by TMT in the hippocampus. Methods: Sixty male Wistar rats were divided into five groups of 12 each. Gro...
