چکیده

There is an obvious need for a technique whereby the medium surrounding an organism under microscopy can be altered without displacing the organism from the field. For example, we are interested in observing the same organisms before, during, and after exposure to various agents. Our general solution is to confine the organisms within a microchamber separated from the well of a well-slide by a semipermeable membrane through which the concentration of convenient reference if observations are to be made in more than one chamber. Input and output channels are ground into a Fisher-Littmad well-slide; this slide is only 1.2 mm thick, and the well has an optically flat bottom. If the grin is fastened flush with the underside of a no. 1 cover slip, there is no problem with standard 97 X objectives. Depending upon the microscope, it may be necessary to change the condenser; the photomicrographs shown here were taken

An Improved Chamber for Direct Visualisation of Chemotaxis

There has been a growing appreciation over the last decade that chemotaxis plays an important role in cancer migration, invasion and metastasis. Research into the field of cancer cell chemotaxis is still in its infancy and traditional investigative tools have been developed with other cell types and purposes in mind. Direct visualisation chambers are considered the gold standard for investigati...

Performances of high numerical aperture water and oil immersion objective in deep-tissue, multi-photon microscopic imaging of excised human skin.

Multi-photon fluorescence microscopy (MPFM) is a powerful technique for imaging scattering, biological specimens in depth. In addition to the sectioning effect generated by the point-like excitation volume, the near-infrared wavelengths used for multi-photon excitation allow deeper penetration into optically turbid specimens. In physiological specimens, the optical properties such as the scatte...

Preparing sample chambers for single-molecule FRET.

Single-molecule (sm) fluorescence detection is a powerful method for studying biological events without time and population averaging. Förster (fluorescence) resonance energy transfer (FRET) is a spectroscopic technique in which the efficiency of energy transfer from donor to acceptor molecules is used to determine distances between molecules in the 30-80 Å range. Structural changes in biologic...

Anti-corrosive Properties of CeO2 Conversion Coatings on Aluminum Alloy 2024 Prepared by Dip Immersion Method

As an alternative to chromate conversion coatings, rare-earth coatings especially cerium, because of the low toxicity, have attracted considerable attention. Using dip immersion method, cerium-based conversion coating was deposited on aluminum 2024. Corrosion resistance was studied in 3.5 wt.% NaCl solution using potentiodynamic polarization, electrochemical impedance spectroscopy, and surface ...

Miicrofluidic-based Oil-immersion Lenses for High Resolution Microscopy

Optical imaging systems with submicron resolution typically require the use of expensive, high numerical aperture (NA) microscope objective lenses. Here, we describe a novel Microfluidic-based Oil-Immersion Lens (μOIL) chip that can perform submicron and wide field of view imaging. Key element of the chip design is the use of high index of refraction, oil-immersion ball mini-lenses that have hi...
