Characterization of Yarrowia lipolytica and related species for citric acid production from glycerol
نویسندگان
چکیده
Twenty-seven Yarrowia lipolytica strains as well as five strains from three other species of the Yarrowia clade (Aciculoconidium aculeatum, andida hispaniensis and Candida bentonensis) were screened for citric acid production with pure glycerol as the carbon source. The cultures were rown under nitrogen-limited conditions. None of the non-Yarrowia strains produced citric acid, although they were able to grow on glycerol. All f the Y. lipolytica strains were able to produce citric acid in varying concentrations and, under the screening conditions used, the yields obtained ormed a continuum from very low to among the highest reported for this substrate. The highest yielding strain, Y. lipolytica NRRL YB-423, roduced 21.6 g/L citric acid from 40 g/L glycerol (54% yield). The citric acid to isocitric acid ratio produced by this strain in the initial screen was 1.3, while most of the strains produced ratios of between 2 and 6. Further work on medium optimization with this strain showed that the optimum arbon to nitrogen ratio for the rate of citric acid production was 172 while the best combination of rate and yield was obtained at a C/N ratio of 43. The citric acid to isocitric acid ratios produced reached an optimum at C/N ratios of 343–686. ublished by Elsevier Inc.

Citric Acid Production by Yeast Grown on Glycerol-Containing Waste from Biodiesel Industry
The possibility of using glycerol and glycerol-containing waste from biodiesel manufacture as a carbon and energy source for microbiological production of citric acid has been studied. Acid formation on the selective media had previously been tested in 66 yeast strains of different genera (Candida, Pichia, Saccharomyces, Torulopsis and Yarrowia). Under growth limitation by nitrogen, 41 strains ...
متن کاملMetabolic Flexibility of Yarrowia lipolytica Growing on Glycerol
The yeast Yarrowia lipolytica is a fascinating microorganism with an amazing metabolic flexibility. This yeast grows very well on a wide variety of carbon sources from alkanes over lipids, to sugars and glycerol. Y. lipolytica accumulates a wide array of industrially relevant metabolites. It is very tolerant to many environmental factors, above all the pH value. It grows perfectly well over a w...
متن کاملA comparative study on glycerol metabolism to erythritol and citric acid in Yarrowia lipolytica yeast cells.
Citric acid and erythritol biosynthesis from pure and crude glycerol by three acetate-negative mutants of Yarrowia lipolytica yeast was investigated in batch cultures in a wide pH range (3.0-6.5). Citric acid biosynthesis was the most effective at pH 5.0-5.5 in the case of Wratislavia 1.31 and Wratislavia AWG7. With a decreasing pH value, the direction of biosynthesis changed into erythritol sy...

متن کاملSubstrates and oxygen dependent citric acid production by Yarrowia lipolytica: insights through transcriptome and fluxome analyses
BACKGROUND Unlike the well-studied backer yeast where catabolite repression represents a burden for mixed substrate fermentation, Yarrowia lipolytica, an oleaginous yeast, is recognized for its potential to produce single cell oils and citric acid from different feedstocks. These versatilities of Y. lipolytica with regards to substrate utilization make it an attractive host for biorefinery appl...
