First identification of polymorphic microsatellite markers in the Burgundy truffle, Tuber aestivum (Tuberaceae)1

چکیده
UNLABELLED PREMISE OF THE STUDY Tuber aestivum, the most common truffle in Europe, plays an important role in the commercial truffle market. For the first time, microsatellite primers were developed to investigate polymorphism within this species. • METHODS AND RESULTS Using direct shotgun pyrosequencing, 15 polymorphic microsatellites were identified out of the 7784 perfect microsatellites present in the 534620 reads obtained. Tested on 75 samples, these microsatellites were highly polymorphic. The number of alleles varied from four to 15, and the expected heterozygosity ranged from 0.266 to 0.620. A multilocus analysis allowed the identification of 63 genotypes over the 75 samples analyzed. • CONCLUSIONS Direct shotgun pyrosequencing is a fast and relatively low-cost technique allowing identification of microsatellites in nonmodel species. The microsatellites developed in this study will be useful in population genetic studies to infer the evolutionary history of this species.

T two best candidate species for truffle cultivation in the south-central U.S. are the Burgundy truffle (Tuber aestivum Vitt., syn. T. uncinatum Ch., Fig. 1a) and the Périgord black truffle (T. melanosporum Vitt., Fig. 1b). These common names (Burgundy truffle and Périgord black truffle) are derived from the names of two of the many regions in France where they are famous. Though native to Euro...
متن کاملNew Insights into the Complex Relationship between Weight and Maturity of Burgundy Truffles (Tuber aestivum)
Despite an increasing demand for Burgundy truffles (Tuber aestivum), gaps remain in our understanding of the fungus' overall lifecycle and ecology. Here, we compile evidence from three independent surveys in Hungary and Switzerland. First, we measured the weight and maturity of 2,656 T. aestivum fruit bodies from a three-day harvest in August 2014 in a highly productive orchard in Hungary. All ...

Of the ten species of European truffle (fungi of the genus Tuber, phylum Ascomycota), some have economic value because of their organoleptic properties (taste and perfume), in particular the black truffle (Tuber melanosporum Vitt.) and the summer and burgundy truffles"2. The black truffle is mainly found in Spain, France and Italy (Fig. la), and it shows variation in several traits, including i...
متن کاملHistorical Biogeography and Diversification of Truffles in the Tuberaceae and Their Newly Identified Southern Hemisphere Sister Lineage
Truffles have evolved from epigeous (aboveground) ancestors in nearly every major lineage of fleshy fungi. Because accelerated rates of morphological evolution accompany the transition to the truffle form, closely related epigeous ancestors remain unknown for most truffle lineages. This is the case for the quintessential truffle genus Tuber, which includes species with socio-economic importance...
متن کاملAssessing genetic diversity of promising wheat (Triticum aestivum L.) lines using microsatellite markers linked with salinity tolerance
Narrow genetic variability may lead to genetic vulnerability of field crops against biotic and abiotic stresses which can cause yield reduction. In this study a set of 37 wheat microsatellite markers linked with identified QTLs for salinity tolerance were used for the assessment of genetic diversity for salinity in 30 promising lines of hexaploid bread wheat (Triticum aestivum L.). A total of 4...
