Selection of Benign Primitive Hematopoietic Progenitors in Chronic Myelogenous Leukemia on the Basis

چکیده
Chronic myelogenous leukemia (CML) is a lethal malignancy of the human hematopoietic stem cell. Here we report that coexistent benign, primitive hematopoietic progenitors can be distinguished from their malignant counterparts in CML bone marrow by differences in cell surface antigen expression. Selection of bone marrow cells expressing the CD34 antigen but lacking the HLA-DR antigen results in recovery of small lymphocyte-like blasts, which initiate and sustain production of myeloid clonogenic progeny in vitro. Secondary clonogenic cells derived at week 1, 5, and 8 from long-term bone marrow cultures (LTBMCs) initiated with

متن کاملMechanisms Underlying Abnormal Trafficking of Malignant Progenitors in Chronic Myelogenous Leukemia
Introduction We studied the adhesion of primitive and committed progenitors from chronic myelogenous leukemia (CML) and normal bone marrow to stroma and to several extracellular matrix components. In contrast to benign primitive progenitors fromCML or normal bone marrow, Ph'-positive primitive progenitors fromCML bone marrow fail to adhere to normal stromal layers and to fibronectin and its pro...
متن کاملPregnancy Outcome of Two Patients with Chronic Myelogenous Leukemia Treated with Imatinib
Although chronic myelogenous leukemia in pregnancy is rare, its management and treatment is more difficult and complicated.Treatment of patients with chronic myelogenous leukemia includes bone marrow transplantation, however in less than 30% of patients the donor’s organ would be accepted. To this end, cytotoxic therapy is considered as an alternative therapeutic option. This option provides sa...
متن کاملBCR-tyrosine 177 plays an essential role in Ras and Akt activation and in human hematopoietic progenitor transformation in chronic myelogenous leukemia.
Chronic myelogenous leukemia (CML) results from the transformation of a primitive hematopoietic cell by the BCR/ABL gene. BCR/ABL signaling has been studied in cell lines and murine models, but the transforming effects of BCR/ABL are highly dependent on cellular context, and mechanisms responsible for the transformation of primitive human hematopoietic cells remain poorly understood. Current ta...
