Leukemia - Associated Changes Identified by Quantitative Flow Cytometry . IV . CD 34 Overexpression in Acute Myelogenous Leukemia

چکیده
During the immunodiagnosis of 517 cases of acute myelogenous leukemia (AML) entered into the Medical Research Council (MRC) AMLIO trials, we have observed the CD34 precursor cell antigen more frequently in AML of M2 morphology, especially in the 84% of cases with the t(8;21) chromosomal translocation, than in any other French-AmericanBritish classification group. CD34 expression was then quantified (using QlFl and Quantum Simply Cellular beads [Flow Cytometry Standards, Research Triangle Park, NCI and CD34+ standard cells). When CD34 antibody-binding capacity (ABC) of normal bone marrow (BM) precursors and leukemic blasts was compared, it was shown that AML M2 cases with t(8;21) not only had the highest percentages of CD34+ blasts, but in >80% of CD34+ cases the individual blasts expressed higher than normal levels of CD34 antigen (>60 x I O 3 ABC per cell). In addition, in 73% of this group CD34 antigen was overexpressed in an asynchronous combination with cytoplasmic myeloperoxidase (MPO). Other signs of asynchrony included high CD34 expression with CD15 andl

Overexpression of MiR-138 Inhibits Cell Growth and Induces Caspase-mediated Apoptosis in Acute Promyelocytic Leukemia Cell Line
Dysregulated expression of miRNAs can play a vital role in pathogenesis of leukemia. The shortened telomere length, and elevated telomerase activity in acute promyelocytic leukemia cells are mainly indicative of extensive proliferative activity. This study aimed to investigate the effect of overexpression of miR-138 on telomerase activity, and cell proliferation of acute promyelocytic leukemia ...

Background & Objective: Acute lymphoblastic leukemia (ALL) is a malignant disease that arises from various mutations in B or T-lymphoid progenitors. MicroRNAs (miRNAs) regulate gene expression by binding to the 3' untranslated region of protein-coding genes. Dysregulation of miRNA expression m...
متن کاملFlow Cytometry in Chronic Myelogenous Leukemia Blast Crisis
Background: Chronic myelogenous leukemia (CML) is a myeloproliferative disorder due to BCR-ABL1 translocation. Patients showing transformation into blast crisis (BC) have poorer treatment response and prognosis. Methods: A retrospective study was conducted in the department of Pathology, Maulana Azad Medical College, New Delhi, India, over a time period of 5 years (2014-2019) to evaluate the ...
متن کاملFamilial Acute Myelogenous Leukemia: Report of Three Cases
Acute myelogenous leukemia (AML) is a hematological malignancy, which accounts for about 15-25% of childhood's leukemia. Genetic factor is one of the most important predisposing elements in childhood acute leukemia, especially AML. In this case report, a rare presentation of familial AML is presented in three monozygotic triplets. Two were 10 months old, and the other one was 16 months old at p...
متن کاملHigh-resolution Antibody Array Analysis of Childhood Acute Leukemia Cells.
Acute leukemia is a disease pathologically manifested at both genomic and proteomic levels. Molecular genetic technologies are currently widely used in clinical research. In contrast, sensitive and high-throughput proteomic techniques for performing protein analyses in patient samples are still lacking. Here, we used a technology based on size exclusion chromatography followed by immunoprecipit...
