Regulation of ß-Galactosidase Synthesis in Wild Type and in a Succinate-Resistant Mutant of Rhizobium meliloti
نویسندگان
چکیده
The synthesis o f /?-galactosidase in Rhizobium meliloti W U60 was found to be inducible by lactose and its non-metabolizable analogue, isopropyl-ß-D-thiogalactoside (IPTG). In contrast to Escherichia coli, galactose and melibiose were very weak inducers o f this enzyme in R. meliloti. The maximum level o f /3-galactosidase in this bacterium is 2% o f that in fully induced E. coli. In addition to glucose, the induced synthesis o f this enzyme in R. meliloti was repressed by galactose, glycerol, and succinate. In comparison to E. coli, addition o f cyclic AMP to the growth medium o f R. meliloti did not alleviate the repressive effect o f the above compounds on ßgalactosidase synthesis. High concentrations o f sodium succinate (100 m M ) were inhibitory to the growth o f R. meliloti. Spontaneous succinate-resistant mutants could be isolated at low frequency. In contrast to the wild type parent, in a succinate-resistant mutant, the synthesis o f /J-galactosidase was not repressed by succinate.

Requirement of succinate dehydrogenase activity for symbiotic bacteroid differentiation of Rhizobium meliloti in alfalfa nodules.
Transmission electron microscopy was used to study the cellular morphologies of a wild-type Rhizobium meliloti strain (L5-30), a nitrogen fixation-ineffective (Fix-) succinate dehydrogenase mutant (Sdh-) strain, and a Fix+ Sdh+ revertant strain within alfalfa nodules and after free-living growth in a minimal medium containing 27 mM mannitol plus 20 mM succinate. The results showed a requirement...

The gene (dctA) encoding the aerobic C(4)-dicarboxylate transporter (DctA) of Escherichia coli was previously mapped to the 79-min region of the linkage map. The nucleotide sequence of this region reveals two candidates for the dctA gene: f428 at 79.3 min and the o157a-o424-o328 (or orfQMP) operon at 79.9 min. The f428 gene encodes a homologue of the Sinorhizobium meliloti and Rhizobium legumin...

The structural requirements for the inhibitor activity of various furan carboxanilide derivatives against succinate dehydrogenase complex (SDC) activity in mitochondria of either wild or mutant strains of Ustilago maydis were investigated with the aid of Hansch QSAR analysis. It has been found that the inhibitor activity against both types of enzymes is best related to the ??? or ??M of th...

An acid-sensitive mutant, TG5-46, derived from Rhizobium meliloti WSM419 by Tn5 mutagenesis, fails to grow below pH 6.0 whereas the parent strain grows at pH 5.7. The DNA sequence of a 2.2 kb rhizobial DNA region flanking Tn5 in TG5-46 contains two open reading frames, ORF1 (designated actS) and ORF2 (designated actR), having high similarity to the sensor-regulator pairs of the two-component sy...
متن کاملRegulation of exopolysaccharide production in Rhizobium leguminosarum biovar viciae WSM710 involves exoR.
A mildly acid-sensitive mutant of Rhizobium leguminosarum bv. viciae WSM710 (WR6-35) produced colonies which were more mucoid in phenotype than the wild-type. Strain WR6-35 contained a single copy of Tn5 and the observed mucoid phenotype, acid sensitivity and Tn5-induced kanamycin resistance were 100% co-transducible using phage RL38. WR6-35 produced threefold more exopolysaccharide (EPS) than ...
