DNA repair: A link between folate deficiency and neuronal cell death

چکیده

Folate is in the core of one-carbon metabolism (OCM), serving as raw material for this biosynthetic pathway essential in DNA synthesis, repair and diverse methylation reactions. Dietary folate is the only source for mammalian OCM requirements and variations in its levels affect intermediary molecules such as the amino acid homocysteine, which is known to have toxic effects at high concentrations. Alterations in OCM is thought to promote apoptosis in nervous system by different ways (i) increasing levels of extracellular homocysteine, what further excitotoxicity in neurons |1,2|; (ii) altering methylation |3| and (iii) influencing the DNA synthesis through precursor imbalance |4|.

Morphological Identification of Cell Death in Dorsal Root Ganglion Neurons Following Peripheral Nerve injury and repair in adult rat

Background: Axotomy causes sensory neuronal loss. Reconnection of proximal and distal nerve ends by surgical repair improves neuronal survival. It is important to know the morphology of primary sensory neurons after the surgical repair of their peripheral processes. Methods: Animals (male Wistar rats) were exposed to models of sciatic nerve transection, direct epineurial suture repair of sciati...

Folate deficiency and ionizing radiation cause DNA breaks in primary human lymphocytes: a comparison.

DNA double-strand breaks, the most serious DNA lesion caused by ionizing radiation, are also caused by several vitamin or mineral deficiencies, such as for folate. Primary human lymphocytes were either irradiated or cultured at different levels of folate deficiency to assess cell proliferation, apoptosis, cell cycle, DNA breaks, and changes in gene expression. Both radiation and folate deficien...

Effects of folate deficiency on gene expression in the apoptosis and cancer pathways in colon cancer cells.

Folate is a B vitamin, deficiency of which appears to increase the risk of developing several malignancies including colorectal cancer. In contrast to the cancer-promoting effect of folate deficiency in normal tissues, several lines of evidence indicate that folate depletion suppresses the progression of existing neoplasms and enhance the sensitivity of cancer cells to chemotherapy. Folate medi...

Folic acid deficiency and homocysteine impair DNA repair in hippocampal neurons and sensitize them to amyloid toxicity in experimental models of Alzheimer's disease.

Recent epidemiological and clinical data suggest that persons with low folic acid levels and elevated homocysteine levels are at increased risk of Alzheimer's disease (AD), but the underlying mechanism is unknown. We tested the hypothesis that impaired one-carbon metabolism resulting from folic acid deficiency and high homocysteine levels promotes accumulation of DNA damage and sensitizes neuro...

Super-Resolution Localization Microscopy of γ-H2AX and Heterochromatin after Folate Deficiency

Folate is an essential water-soluble vitamin in food and nutrition supplements. As a one-carbon source, it is involved in many central regulatory processes, such as DNA, RNA, and protein methylation as well as DNA synthesis and repair. Deficiency in folate is considered to be associated with an increased incidence of several malignancies, including cervical cancer that is etiologically linked t...
