چکیده

Catharanthus roseus (L.) G. Don. accumulates in the leaves the dimeric terpenoid indole alkaloids (TIAs) vinblastine and vincristine, which were the first natural anticancer products to be clinically used, and are still among the most valuable agents used in cancer chemotherapy. The great pharmacological importance of the TIAs, associated with the low abundance of the anticancer alkaloids in the plant, stimulated intense research on the TIA pathway, and C. roseus has now become one of the most extensively studied medicinal plants. About 130 TIAs have already been isolated from C. roseus, and it has been shown that the biosynthesis of vinblastine is highly complex, involving at least 30 steps from the amino-acid tryptophan and the monoterpenoid geraniol (Loyola-Vargas et al., 2007; van der Heijden et al., 2004). All the TIAs of C. roseus derive from the common precursor strictosidine, after which the TIA pathway splits into several branches including a short one leading to ajmalicine and serpentine (used as an antihypertensive and as a sedative respectively), and two long branches leading to vindoline and catharanthine the leaf abundant monomeric precursors of vinblastine and vincristine (Loyola-Vargas et al., 2007; van der Heijden et al., 2004). In our lab, we have performed the characterization of a key biosynthetic step of the anticancer TIAs the biosynthesis of the first dimeric TIA, α-3’,4’-anhydrovinblastine (AVLB), from vindoline and catharanthine. We identified a leaf class III peroxidase (Prx) with AVLB synthase activity and we purified and characterized this enzyme, which was named Catharanthus roseus peroxidase 1, CroPrx1 (Bakalovic et al., 2006; Peroxibase, http://peroxibase.toulouse.inra.fr index.php). We have further shown the localization of CroPrx1 in the same subcellular compartment where alkaloids accumulate, the vacuole, through biochemical and molecular methodologies,

The expression profile of D4H and DAT genes in Catharanthus roseus in response to drought, salinity and salicylic acid

Catharanthus roseus L. is an important medicinal plant producing several terpenoid indole alkaloids (TIAs) such as vincristine and vinblastine secondary metabolites with anticancer activity. The TIAs biosynthetic pathways are affected by biotic and abiotic stresses. In this study the effect of drought (7 days), salinity (150 mM NaCl), foliar spray of salicylic acid (10-5 M) and salicylic acid i...

PLANT REGENERATION FROM MESOPHYLL PROTOPLASTS OF POTATO (SOLANUM TUBEROSUM L.) CULTIVAR DELAWARE USING SILVER THIOSULFATE (STS)

The use of in vitro potato shoot cultures for protoplast isolation is desirable for consistent protoplast qualities. Ethylene build-up in the culture vessels causes problems for leaf growth (e.g. decreasing leaf surface, dry weight and chlorophyll) in shoot culture. Fifty to one hundred µM Silver thiosulfate (STS) produced a larger leaf area as a source of plant material for protoplast isolatio...

A Novel AP2/ERF Transcription Factor CR1 Regulates the Accumulation of Vindoline and Serpentine in Catharanthus roseus

As one type of the most important alkaloids in the world, terpenoid indole alkaloids (TIAs) show a wide range of pharmaceutical activities that are beneficial for clinical treatments. Catharanthus roseus produces approximately 130 identified TIAs and is considered to be a model plant to study TIA biosynthesis. In order to increase the production of high medical value metabolites whose yields ar...

Improving Effects of Mild Cold Stress and Salicylic acid on Growth and Physiology of Periwinkle (Catharanthus roseus Don.)

To study effects of salicylic acid (SA) treatments (0, 0.5, 1 and 2 mM) in two forms of seed priming (for 24 h) and spraying on seedling at 4-6 leaf stage an experiment was conducted on periwinkle (Catharanthus roseus Don.) with and without exposure to a mild cold stress of 8 °C (for 32 h). Seeds were sown in trays containing peat-based substrate (with 20% sandy loam field soil). Transplants of...

Mutation breeding in Catharanthus roseus (L.) G. Don: An Overview

Induced mutagenesis is best approach for creation of genetic variability but most of interest is paid towards crop plants with only a few exceptions. But even widely used medicinal plants could receive a little attention in this direction. Catharanthus roseus is an important plant medicinal plant yielding antihypertensive alkaloid ajmalcine and anticancerous alkaloid vincristine and vinblastine...
